摘要
考察不同型号大孔树脂对罗田金银花叶中绿原酸的分离效果,为进一步开发绿原酸相关产品提供参考。采用静态和动态吸附法考察大孔树脂的吸附、解吸和分离效果;采用紫外分光光度法测定绿原酸含量。结果表明:D101树脂综合性能最佳,分离纯化金银花叶提取液中绿原酸的最佳工艺条件为树脂柱高径比3,上样液浓度0.8mg/mL,pH 4,去离子水洗脱流速4BV/h,用量5倍柱体积。所得提取物中绿原酸含量为28.21%。D101大孔树脂对金银花叶中绿原酸分离效果好,工艺经济可行,可用于工业化生产。
In order to develop products about Flos Lonicerae further,the isolation of chlorogenic acid from leaves of Luotian honeysuckle with different macroporous resins were investigated.The adsorption,desorption and separation were investigated by static and dynamic adsorption methods;the content of chlorogenic acid was determined by UV spectrophotometry.Results: The macroporous resin of D101 had the best effect.The optimum separation conditions were as follows: height to diameter ratio of resin column,3;sample concentration,0.8 mg/mL;pH,4;elution flow rate,4 BV/h;elution volume,5 BV.Under such conditions,the content of chlorogenic acid in extraction is 28.21%.The macroporous resin of D101 is a choice of the optimal macroporous resin to isolate chlorogenic acid from leaves of Luotian honeysuckle.This method is practical and reasonable,and suitable for commercial production.
