摘要
目的建立高效液相色谱(HPLC)法测定复方氯己定凝胶中醋酸氯己定和盐酸达克罗宁的含量。方法选用Inertsil ODS-SP色谱柱,0.5%三乙胺(用冰醋酸调pH值至3.8)-乙腈(68∶32)为流动相,流速1.0 mL/min,检测波长270 nm。结果醋酸氯己定在0.081 52~0.489 10μg时呈良好的线性关系(r=0.999 9,n=6),平均回收率为98.82%,相对标准偏差(RSD)为1.19%(n=9);盐酸达克罗宁在0.163 6~0.981 6μg时呈良好的线性关系(r=0.999 9,n=6),平均回收率为98.85%,RSD为1.13%(n=9)。结论 HPLC法操作简便、灵敏度高、重复性好,可用于该制剂中醋酸氯己定和盐酸达克罗宁的含量测定。
Objective To establish a HPLC method for determining the content of chlorhexidine acetate and dyclonine hydrochloride in Compound Chlorhexidine Gel.Methods The Inertsil ODS-SP chromatographic column was used with 0.5% triethylamine(adjusted pH to 3.8 with acetic acid)-acetonitrile(68∶32) as the mobile phase.The flow rate was 1.0 mL/min and detecting wavelength was 270 nm.Results The concentration of chlorhexidine acetate in the range of 0.08152-0.4891 μg showed a good linear relationship(r=0.999 9,n=6),the average recovery rate was 98.82%,RSD 1.19%;the concentration of dyclonine hydrochloride in the range of 0.1636-0.9816 μg showed a good linear relationship(r=0.999 9,n=6),the average recovery rate was 98.85%,RSD 1.13%(n=9).Conclusion The HPLC method is simple to operate and highly sensitive with good repeatability,and can be used for the content determination of chlorhexidine acetate and dyclonine hydrochloride in this preparation.
