摘要
目的 :探讨星形细胞在慢性脑灌注不足脑损害中的作用。方法 :70只老龄大鼠持久性双侧颈总动脉结扎 (2VO) ,其中 12只接受环孢霉素A(CsA)胃灌治疗。免疫组化法多克隆抗血清GFAP标记星形细胞。实验研究为持久性 2VO 1~ 4月。结果 :持久性 2VO 1月皮层、海马和白质处的星形细胞明显增生、肥大。 1~ 4月 ,皮层、海马和白质处星形细胞继续增生、肥大 ,同时脑损害加重。CsA治疗后脑损害明显减轻 ,星形细胞的活动明显减少。结论 :大鼠慢性脑灌注不足可诱导星形细胞持久性增生、肥大 ,反应性星形细胞扮演神经保护作用。CsA可通过减轻脑损害 ,从而抑制大鼠慢性脑灌注不足脑内星形细胞的活动。
Objective:To investigate the effects of activation of astrocytes on brain damages after chronic cerebral hypoperfusion.[WT5HZ]Methods:Both common carotid arteries were ligated bilaterally in 70 Wistar rats.Twelve of them were filled cyclosporin A.Astrocytes were observed with immunohistochemistry for GFAP.Activation of astrocytes and brain damages were then investigated from 1 to 4 months after the ligation.[WT5HZ]Results:There were obvious proliferation and hypertrophy of astrocytes in cortex,hippocampus and white matter after chronic cerebral hypoperfusion.From 1 to 4 months, proliferation and hypertrophy of astrocytes were continued.Meanwhile,brain damages have become heavier,In cyclosporin A-treated rats,brain damages were much less intense,proliferation and hypertrophy of astrocytes were significantly reduced also.[WT5HZ]Conclusion:Proliferation and hypertrophy of astrocytes were induced in chronic cerebral hypoperfusion,reactive astrocytes displayed a neuroprotected pattern,Cyclosporin A suppressed activation of astrocytes by reduced brain damages after chronic cerebral hypoperfusion.
