摘要
1目的 探讨尼莫通对大鼠脑缺血再灌注时细胞凋亡的影响。 2方法 对实验大鼠于脑缺血前应用尼莫通进行干预 ,在脑缺血再灌注不同时间内 ,用免疫组化法检测凋亡相关基因 c- fos及 bcl- 2蛋白表达的变化。 3结果 尼莫通组大鼠脑缺血后的梗死体积显著减小 (t=2 .79,P<0 .0 5 ) ;尼莫通组脑缺血再灌注时皮层和基底核区 c- fos的表达明显下降 (t=4.84,6 .14,P<0 .0 0 1) ,而 bcl- 2蛋白的表达明显增强 (t=3.5 3,4.2 2 ,P<0 .0 0 1)。 4结论 尼莫通可抑制脑缺血再灌注时细胞凋亡的发生。
Objective To investigate the effect of nimotop(NM) on apoptosis after focal cerebral ischemia followed by reperfusion and thus provide more theoretical basis for clinical application of nimotop for the treatment of ischemic cerebrovascular diseases. Methods Rats were pretreated with NM(500μg/kg) 30min prior to MCAO,changes of protein expression of c fos and bcl 2 in rats after ischemia and reperfusion were observed by immunohistochemical method. Results The infarct volume in NM group was significantly smaller than that in the corresponding regions in the control group(t=2.79,P<0.05).In NM group,the protein expression of c fos decreased significantly after ischemia/reperfusion both in the ischemic cortex and in the ischemic basal ganglia(t=4.84,6.14,P<0.001),but the protein expression of bcl 2 increased significantly after ischemia/reperfusion both in the ischemic cortex and in the ischemic basal ganglia (t=3.53,4.22, P<0.001). Conclusion NM may inhibit the development of apoptosis in rats after ischemia and reperfusion and have the neuro protective effect against ischemia. [
