多西环素与地塞米松抑制碱烧伤大鼠角膜新生血管的对比研究 被引量:3

Comparison of inhibitory effects of Doxycycline and Dexamethasone on corneal neovascularization in rats with corneal alkali burn

摘要 目的对比多西环素、地塞米松对碱烧伤大鼠角膜新生血管的抑制作用。方法健康SD大鼠48只,随机分为对照组、多西环素组、地塞米松组,每组各16只。建立角膜碱烧伤模型后,分别予眼液溶媒、5g·L-1多西环素眼液、1g·L-1地塞米松眼液点眼,每天4次。于碱烧伤后3d、7d、14d、21d裂隙灯显微镜下观察,计算炎症指数和角膜新生血管面积。结果 3d、7d多西环素组、地塞米松组结膜充血、角膜水肿较对照组轻,炎症指数多西环素组分别为0.45±0.03、0.22±0.14,地塞米松组分别为0.50±0.14、0.22±0.07,炎症指数均明显低于对照组(均为P<0.05),多西环素组、地塞米松组间差异无统计学意义(P>0.05);14d、21d地塞米松组炎症指数分别为0.44±0.30、0.47±0.19,显著高于多西环素组(0.04±0.06、0.19±0.11),差异均有统计学意义(均为P<0.05)。多西环素组、地塞米松组各时间点角膜新生血管面积小于对照组(均为P<0.05),3d、7d地塞米松组角膜新生血管面积小于多西环素组(均为P<0.05),14d、21d两组比较差异均无统计学意义(均为P>0.05)。地塞米松组前房出血12.5%,对照组为43.8%,多西环素组为6.3%。对照组角膜溃疡5眼,地塞米松组为8眼,多西环素组为1眼。除多西环素组无一眼发生眼内炎外,余两组均有眼内炎发生。结论碱烧伤后短期内地塞米松抑制大鼠角膜新生血管的作用强于多西环素,但长期效果后两者无显著差异。 Objective To compare the inhibitory effects of Doxycycline and Dex- amethasone on corneal neovascularization (CNV) in rats with corneal alkali burn. Methods A total of 48 SD normal rats were randomly and evenly divided into control group, Doxycycline-treated group and Dexamethasone-tmated group. After the alkali- burnt models were built, they were treated with ocular humor menstruum, 5 g ~ L-1 Doxycycline and l g ~ L-' Dexamethasone, respectively, 4 times a day. They were ob- served at postoperative 3 days,7 days,14 days and 21 days by slitlamp microscope,and inflammatory index and area of CNV. Results The conjunctival congestion and corneal edema of Doxycycline-treated group and Dexamethasone-tmated group were lighter than those of control group at postoperative 3 days and 7 days, the inflammatory indexes of Doxycycline-tmated group (0. 45 ± 0. 03 and 0. 22± 0. 14,respectively) and Dexametha- sone-tmated group(0.50 ±0. 14 and 0. 22± 0.07, respectively) were lower than those of control group( all P 〈0.05 ) ,and there was no significant difference between Doxycycline- treated group and Dexamethasone-treated group. At postoperative 14 days and 21 days, the inflammatory indexes of Dexamethasone-tmated group(0.44 ±0.30 and 0.47 ±0.19, respectively) were significantly higher than those of Doxycycline-treated group (0.04 ± 0. 06 and 0.19 ± 0. 11, respectively) ( both P 〈 0.05 ). At all time points,the areas of CNV in both Dexamethasone-tmated group and Doxycycline-tmated group were smaller than those in control group ( both P 〈 0.05 ) ; at postoperative 3 days and 7 days, the areas of CNV in Dexamethasone-treated group were smaller than in Doxycycline-treated group (both P 〈0.05) ;at postoperative 14 days and 21 days,them was no significant difference between Dexamethasone-treated group and Doxycycline-treated group ( both P 〉 0. 05 ). There was hyphema in 12.5% of Dexamethasone-tmated group,43.8% of control group and 6.3% of Doxycycline-treated group. There was corneal ulcer in 5 eyes of control group,8 eyes of Dexamethasone-treated group and 1 eye of Doxycycline-treated group. The endophthalmitis only appeared in Dexamethasone-treated group and control group. Conclusion The inhibitory effect of Dexamethasone on CNV in rats with corneal alkali bum is stronger than Doxycycline in short term,while in long term there is no difference between them.
