β-半乳糖苷酶产生菌的筛选和发酵条件优化 被引量:3

摘要 以X-gal为显色剂,从大连三寰乳制品厂区周围土壤中筛选到15株β-半乳糖苷酶产生菌,以ONPG为底物,测定该15株菌液体发酵后的β-半乳糖苷酶活力,复筛到1株β-半乳糖苷酶高产菌株MFY-03。对菌株MFY-03摇瓶发酵产β-半乳糖苷酶的工艺条件进行了优化,确定了发酵培养基的组成为:葡萄糖1.0%,乳糖1.0%,酵母粉0.5%,蛋白胨1.0%,吐温80 1.0%,pH 6.8,于45℃、200 r/min条件下培养24 h,β-半乳糖苷酶活力达到3.74 U/mL。 Fifteen strains which produce β-galactosidase were screened from soil of Dalian Sanhuan dairy plant using X-gal as indicator.The β-galactosidase activity was measured using ONPG as substrate,and the strain of MFY-03 was selected among the above fifteen strains based on the β-galactosidase activity.The fermentation conditions for β-galactosidase production by the strain MFY-03 in shaking flasks were optimized.The optimal fermentation medium for β-galactosidase production was: 1 % glucose,1 % lactose,0.5 % yeast powder,1 % peptone,1 % Tween-80,and pH 6.8.The flasks were incubated at 45 ℃ and 200 r/min for 24 h,and the activity of β-galactosidase reached 3.74 U/mL.
