大鼠脑出血后细胞色素c和神经细胞凋亡的关系及立体定向血肿抽吸术的干预作用 被引量:1

摘要 目的研究大鼠脑出血(intracerebral hemorrhage,ICH)后细胞色素c(Cytochrome c)表达和神经细胞凋亡的关系,探讨立体定向血肿抽吸术的干预作用及其机制。方法用Ⅳ型胶原酶注入到大鼠右侧苍白球诱导ICH模型。用凋亡原位末端标记技术(TUNEI)和免疫组化方法检测血肿周围神经细胞凋亡和Cyt c表达的动态变化。结果ICH后6 h Cyt c表达开始增加,3 d达高峰,此后逐渐减少,11 d仍可见少量表达。ICH后6 h血肿周边组织可见TUNEL阳性细胞,24 h~3 d达高峰,然后逐渐较少,7 d、11d时仍见较多凋亡细胞。ICH后11 d内Cyt c表达和神经细胞凋亡呈正相关(r=0.808,P<0.01)。立体定向血肿抽吸治疗组Cyt c和TUNEL阳性细胞数较ICH自然恢复组明显减少(P<0.05)。结论ICH后血肿周围神经细胞损伤有凋亡机制参与,Cyt c释放是神经细胞凋亡的一个关键事件,立体定向血肿抽吸术可以有效减轻脑出血后的细胞凋亡,其机制之一是抑制凋亡通路Cyt c的释放。 Objective To observe the relationship of Cytochrome c expression and apoptosis of neuron in rats following intracerebral hemorrhage(ICH), and investigate the effect of hematoma aspiration. Methods ICH models was induced in rats by injection of type IV collagenase into the right caudate nucleus. With TUNEL and immuno- histochemistry, We investigated the dynamic changes of apoptosis and the expression of cytochrome C in the tissue surrounding hematoma in ICH rats. Results Cyt c expression was increased at 6 h, peaked at 3 d and then de- creased, but after lld there were still few posivite cells postoperative. TUNEL- positive cells appeared in the model group 6 h after collagenase injection, peaked at 3 d and then declined, but after 7 d, 11 d there were still many apoptotic cells. There was a positive correlation between Cyt c expression and cell apoptosis in 11 d after ICH (r = 0. 808, P 〈 0.01). Cytochrome c - positive neurons and the apoptotic cells of hematoma aspiration group greatly decreased than those of natural recovery group ( P 〈 0.05). Conclusion Apotosis is a pattern of death after intracerebral hemorrhage, and the release of Cyt c from mitochondria to cytosol is a critical step in ap- optotic cell death after ICH. hematoma aspiration can effectively alleviate apoptosis after ICH by preventing Cyt c l'~l^e.
