摘要
目的:探讨加味茵陈蒿汤对湿热内蕴证大鼠急性肝损伤的治疗作用机制。方法:SPF级SD大鼠,雌性,72只,随机分为空白组、模型组、西药组(复方甘草酸苷)组、中药加味茵陈蒿汤低、中、高剂量组。所有大鼠均适应性喂养在室温15~20℃,相对湿度60%~80%的实验室7 d。7 d后给模型组、西药组、中药组予1次性ip D-氨基半乳糖600 mg.kg-1以造成急性肝损伤模型,造模后除模型组外余各组ig给予药物,每日2次,复方甘草酸苷组剂量为15.75 mg.kg-1,加味茵陈蒿汤低、中、高剂量组剂量分别为14.7,29.4,58.8 g.kg-1,空白组予以0.9%的生理盐水ig,灌药体积每次10 mL.kg-1,2次/d,共ig治疗2 d。造模48 h后断颈处死动物剖取肝脏,RT-PCR和Western blot法检测肝脏细胞Toll样受体4(Toll-like receptor 4,TLR-4)mRNA及其蛋白表达。结果:与正常组比较,模型组大鼠肝脏组织TLR-4mRNA和TLR-4蛋白均有极显著增加(P<0.01);与模型组比较,加味茵陈蒿汤可显著下调TLR-4 mRNA(P<0.01)及TLR-4蛋白(P<0.01)的表达。结论:加味茵陈蒿汤可通过下调肝脏细胞TLR-4 mRNA及TLR-4蛋白表达有效干预肝脏细胞纤维化病变。
Objective: To explore the mechanism of the modified Yinchenhao decoction(YCHT) in treating acute liver injury.Method: Seventy-two female SD rats were randomized into six groups with 12 in each group: blank control group,model group,compound glycyrrhizin tablets group,and low,middle and high dosage group of supplemented Yinchenhao decoction.All the rats were fed with common feed and clean water in the 15-20 ℃ and humidity level at 60%-80% of laboratory for seven days,seven days later,single intraperitoneal injection of D-GalN at a dosage of 600 mg.kg-1 was administered to the rats except those in the blank control group to induce acute liver injury.Medications were given orally to each treatment group at the same time,the daily dose of compound Glycyrrhizin tablets was 15.75 mg.kg-1,and dosage of the three herbal groups was 14.7,29.4,58.8 g.kg-1 respectively.The blank control group was fed with the 0.9% physiological saline.All the medications were given at a volume of 10 mL.kg-1,twice a day for two days.48 h after modeling,all rats were sacrificed and livers werecollected,RT-PCR and western blot were used to detect the expression of TLR-4 mRNA and TLR-4 protein in liver cells.Result: Compared with the model group,YCHT could significantly reduce the TLR-4 mRNA (P 0.01) and TLR-4 protein expression(P 0.01).Conclusion: Through reducing the TLR-4 mRNA and TLR-4 protein expression of rats liver cells,modified YCHT could effectively intervene the liver cells fibrosis.
