摘要
目的 通过检测食管鳞癌光动力治疗前后凋亡相关调控基因表达的变化,探讨光动力治疗在食管鳞癌中的作用机制.方法 采用免疫组织化学法检测23例食管鳞癌患者(食管鳞癌组)及20例健康体检者(对照组)食管黏膜组织中Bax、Bcl-2、Fas及FasL蛋白的表达.结果 Bax、Fas及FasL蛋白在食管鳞癌组的表达较对照组降低(1.08±0.37比1.41±0.26,0.63±0.47比1.35±0.54,0.82±0.39比1.67±0.68),Bcl-2在食管鳞癌组的表达较对照组上调(2.72±0.31比1.55±0.09),差异均有统计学意义(P<0.05).经光动力治疗后,Bax、Fas及FasL在食管鳞癌组的表达较治疗前上升,差异有统计学意义(P<0.05),Bcl-2在食管鳞癌组的表达较治疗前下调,差异有统计学意义(P<0.05).结论 食管鳞癌细胞中凋亡相关调控基因表达的失调可能与食管鳞癌的发生、发展有关;光动力治疗食管鳞癌的作用机制之一是调节细胞凋亡相关调控基因表达,从而抑制食管鳞癌细胞增殖.
Objective To explore the therapeutic mechanism of photodynamic therapy in esophageal squamous cell carcinoma,by detecting the expression of apoptosis related regulatory gene before and after photodynamic therapy.Methods The immunohistochemical staining was used to detect the expression of Bax,Bcl-2,Fas and FasL protein in 23 cases of esophageal squamous cell carcinoma (esophageal squamous cell carcinoma group) and 20 cases of healthy people (control group).Results The expression of Bax,Fas and FasL in esophageal squamous cell carcinoma group was significantly lower than that in control group (1.08 ± 0.37 vs.1.41 ± 0.26,0.63 ± 0.47 vs.1.35 ± 0.54,0.82 ± 0.39 vs.1.67 ± 0.68,P 〈 0.05).The expression of Bcl-2 in esophageal squamous cell carcinoma group was significantly higher than that in control group (2.72 ±0.31 vs.1.55 ± 0.09,P 〈 0.05).The expression of Bax,Fas and FasL in esophageal squamous cell carcinoma group after photodynamic therapy was significantly higher than that before photodynamic therapy (P 〈 0.05).The expression of Bcl-2 in esophageal squamous cell carcinoma group after photodynamic therapy was significantly lower than that before photodynamic therapy (P 〈 0.05).Conclusions The apoptosis regulatory genes are related to the occurrence,development of esophageal squamous cell carcinoma.One of the mechanisms related to therapeutic effect of PDT is its induction of gene-oriented apoptosis,which can inhabit cell proliferation of the esophageal squamous cell carcinoma.
