益气活血方和补肾生髓方对脑缺血再灌注大鼠Notch3和Frizzled2 mRNA及蛋白表达的影响 被引量:18

摘要 目的:探讨益气活血方和补肾生髓方对脑缺血再灌注大鼠额顶叶皮质Notch3和Frizzled2 mRNA表达及这2种蛋白在海马CA1区表达的影响。方法:线栓法制作局灶性脑缺血再灌注大鼠模型,脑缺血2 h,再灌注1周,随机分为假手术组、缺血再灌注组、益气活血方组和补肾生髓方组。采用Notch信号通路基因芯片PCRAssay检测额顶叶皮质Notch3和Frizzled2 mRNA表达,免疫组化EnVision两步法检测海马CA1区Notch3和Friz-zled2蛋白表达。结果:缺血再灌注组Frizzled2 mRNA表达显著高于假手术组(P<0.05),益气活血方组Notch3mRNA表达显著高于缺血再灌注组(P<0.05),补肾生髓方组Frizzled2 mRNA表达显著低于模型组(P<0.05),益气活血方组Frizzled2 mRNA表达显著高于补肾生髓方组(P<0.05)。缺血再灌注组Notch3和Frizzled2蛋白表达均高于假手术组(P<0.05),2种中药复方组Notch3和Frizzled2蛋白表达均高于缺血再灌注组(P<0.05),补肾生髓方组Notch3蛋白表达高于益气活血方组(P<0.05)。结论:益气活血方和补肾生髓方能通过增强额顶叶或海马CA1区Notch3 mRNA及蛋白表达,降低Frizzled2 mRNA及蛋白表达,调节Notch信号转导通路,促进脑损伤修复。 AIM: To investigate the effects of Yiqi-Huoxue prescription and Bushen-Shengsui prescription on the expression of Notch3 and Frizzled2 at mRNA and protein levels in the cortex of frontal and parietal lobes or hippocampal CA1 area after local cerebral ischemia / reperfusion(IR) in rats.METHODS: The local cerebral IR model was established by intraluminal thread occlusion of the middle cerebral artery(MCAO).The animals were randomly divided into sham operation group(sham group),IR group,Yiqi-Huoxue prescription(IR + Y) group and Bushen-Shengsui prescription(IR + B) group.Using the techniques of PCR and immunohistochemical staining,the expression of Notch3 and Frizzled2 at mRNA and protein levels was observed at the 1st week in the cortex of frontal and parietal lobes or hippocampal CA1 area.RESULTS: The expression of Frizzled2 mRNA in IR group was higher than that in sham group.The expression of Notch3 mRNA in IR + Y group was higher than that in IR group.The expression of Frizzled2 mRNA in IR + B group was lower than that in IR group.The expression of Frizzled2 mRNA in IR + Y group was higher than that in IR + B group in the cortex of frontal and parietal lobes.The protein levels of Notch3 and Frizzled2 in IR group were higher than those in sham group,and those in IR + Y group and IR + B group were higher than those in IR group.The protein level of Notch3 in IR + B group was higher than that in IR + Y group in hippocampal CA1 area.CONCLUSION: Yiqi-Huoxue prescription and Bushen-Shengsui prescription regulate the Notch signal transduction pathway by enhancing Notch3 and inhibiting Frizzled2 at mRNA and protein levels after local cerebral ischemia / reperfusion in MACO rats,thus promoting the repair of brain injury.
