低氧对大鼠睾丸组织死亡受体5和FLIP抑制蛋白表达的影响 被引量:4

摘要 目的:检测常氧与模拟海拔4 000 m低氧条件下大鼠睾丸组织中死亡受体5(DR5)和FLIP抑制蛋白(c-FLIP)表达水平,以及c-FLIP在睾丸组织中的分布。方法:40只10周龄雄性SD大鼠随机分为常氧组,低氧3、15、30 d组。低氧各组分别置模拟海拔4 000 m低压舱中减压3、15、30 d;常氧组在平原(海拔300 m)喂养;减压结束后取睾丸组织,Western印迹检测睾丸组织中DR5与c-FLIP蛋白表达,免疫荧光法检测睾丸组织中c-FLIP分布。结果:低氧3、15、30 d组大鼠睾丸DR5表达量分别为2.04±0.11、1.97±0.12、2.34±0.11,均显著高于常氧组DR5的表达(1.78±0.09,P均<0.05)。低氧15、30 d组大鼠睾丸c-FLIP表达量分别为0.87±0.03、0.74±0.07,均显著低于常氧组c-FLIP的表达(1.03±0.02,P均<0.05)。结论:模拟海拔4 000 m低氧促进睾丸组织凋亡蛋白DR5表达,抑制睾丸组织抗凋亡蛋白c-FLIP表达。 Objective : To evaluate the effects of hypobaric hypoxia on the expressions of death receptor 5 (DRS) and cellular FLICE-like inhibitory protein (c-FLIP) and the distribution of c-FLIP in the rat testis. Methods : Forty adult male SD rats were ran- domly divided into four groups of equal mlmber: normoxia control, 3 d hypoxia, 15 d hypoxia and 30 d hypoxia. The control rats were raised at 300 m above the sea level, while the latter three groups of rats in a hypobaric chamber at a simulated altitude of 4000 m for 5, 15 and 30 days, respectively. Then the expressions of DR5 and c-FLIP were detected by immunoblotting and the distribution of c-FLIP in the testis observed by immunofluorescence. Results : The expressions of DR5 were 2.04 ± 0. 11, 1.97 ± 0. 12 and 2.34 _± 0.11 in the 3 d, 15 d and 30 d hypoxia groups, respectively, significantly higher than 1.78 ±0.09 in the normoxia group (P 〈0.05). The ex- pressions of c-FLIP were 0.87 ±0.03 and 0.74 ±0.07 in the 15 d and 30 d hypoxia groups, respectively, significantly lower than 1.03 ± 0. 02 in the normoxia group (P 〈 0.05). Conclusion : Simulated hypobaric hypoxia at 4000 m above the sea level increasedthe expression of DR5 and inhibited that of c-FLIP in the rat testis.
