摘要
目的:探讨阿魏酸钠对糖尿病大鼠肾脏的保护机制。方法:50只雄性SD大鼠随机分为2组,正常对照组10只,造模组40只。造模组采用空腹一次性尾静脉注射链脲佐菌素(STZ)55mg/kg制造糖尿病模型,72小时后血糖≥16.7mmol/l为造模成功。将造模成功的40只大鼠随机分为糖尿病模型组、阿魏酸钠50mg/kg、100mg/kg、200mg/kg,给药8周后全自动化生化分析仪检测生化指标,免疫组化法和Western Blot法检测转化生长因子-β1(TGF-β1)、Smad1、Ⅳ型胶原(ColⅣ)定位和半定量表达,电镜观察肾脏组织超微结构。结果:与糖尿病模型组相比,阿魏酸钠高中低剂量组基底膜增厚、系膜基质积聚、足突融合均减轻,肾脏组织中TGF-β1、Smad1、ColⅣ表达减少,且有剂量依赖性,高剂量组改变最明显。结论:阿魏酸钠对糖尿病肾脏具有保护作用,可能与降低TGF-β1、Smad1、ColⅣ的表达有关。
Objiective: To study the protection mechanism of sodium ferulate(SF) on diabetic rats' kidney.Methods : 50 male SD rats were randomly allocated into 2 groups: normal control group(n = 10),modle group(n = 40).Rats in the modle group were administered with strepto-zotocin(55mg/kg) by vena caudalis injection to induce diabetic model.The modle diabetes mellitus were considered to be successful when the concentration of blood glucose was constantly ≥16.7mmol/l 72 hours after the injection.The modle rats were divided randomly into 4 groups: diabetes mellitus modle group(n = 10),SF high dosages treatment group(n = 10),SF middle dosages treatment group(n = 10) and SF low dosages treatment group(n = 10).The latter 3 groups were given corresponding dosages of 50mg /(kg.d),100mg /(kg.d) and 200mg /(kg.d).The interference lasted for 8 weeks.Biochemical indices were obtained by automatic analyzer.Immunohistochemical method and Western Blotting analysis were used to detect the protein expression of smad1 and transforming growth factor-β1(TGF-β1),type Ⅳcollagen(Col Ⅳ).The Renal ultrastructure was observed by electron microscope.Results: Comparing with diabetes modle group,SF high,middle and low groups can reduce the expansion of mesangial matrix and thickening of the glomerular basement membrane,downgrade the expression of Smad1,TGF-β1 and ColⅣwith dosages-response relationship.SF high group changes more obvious.Conclusion: SF on diabetic kidney may have a protective effect.The mechanism may relate with reducing the expressing of TGF-β1,Smad1 andColⅣ.
