黄芪对脾虚发热大鼠的作用 被引量:11

Observation of the effective of Astragalus mongholicus on the model of spleen-qi deficiency with fever in rats

摘要 目的:探讨不同剂量黄芪对脾虚发热大鼠模型的作用。方法:大鼠随机分为正常对照组、脾虚发热组、黄芪大、中、小剂量组共5组。采用饮食失节+游泳疲劳制备脾虚模型,实验第18天开始,黄芪各组大鼠分别按剂量2.16g/kg(大)、1.08g/kg(中)、0.54g/kg(小)给予灌胃,正常组和脾虚发热组给予同体积蒸馏水,每日1次,连续5天。第22d的AM 8:00,各实验组大鼠ipLPS(80μg/kg),正常对照组ip等量生理盐水,观测每周各组大鼠的外观行为、LPS注射后不同时间点的体温和血IL-1β、IL-2、GAS、CCK、B-EP、SS。结果:与正常组相比,脾虚发热组大鼠在ip LPS后60、120、180min时的体温出现明显升高(P<0.05),外观行为积分值显著升高,血中SS、IL-1β、IL-2含量见显著升高(P<0.05)。与脾虚发热组比较,黄芪大剂量组外观行为的部分指标有所改善;黄芪三个剂量组60min后的体温均呈现不同程度的降低,其中大、中剂量组的多点体温呈显著降低(P<0.05);大剂量组IL-2和GAS含量均见显著降低(P<0.05),中剂量组SS/CCK含量呈显著降低/升高(P<0.05),小剂量组GAS和SS含量均见显著性降低(P<0.05)。较之于中、小剂量组,黄芪大剂量组IL-2含量降低明显(P<0.05)。结论:黄芪对脾虚发热大鼠模型有不同程度的调节作用,综合效应指标则以黄芪大剂量作用明显。 to investigate the effective by different dosages of Astragalus mongholicus on the model of spleen-qi deficiency with fever in rats. Method: The male wistar rats were assigned into the normal group, the spleen-qi deficiency with fever group, the large dosage of As- tragalus mongholicus group, the medium dosage of Astragalus mongbolicus group, the small dosage of Astragalus mongholicus group. The rats of spleen-qi deficiency with fever group and different dosages of astragalus mongholicus group were treated with excess fatigue plus out of con- stant diet to copy spleen-qi deficiency rat model. From the 18th day, every taken medicine group rats according body surface area were fed with 2.16g/kg ( large), 1.08g/kg ( medium), and 0.54g/kg (small). The rats of the normal group and the spleen-qi deficiency with fever group were fed with physiological saline once a day for 5 days. At 8:00 am on the 22th day,the rats of different dosages of Astragalus mong- holicus group and the spleen-qi deficiency with fever group were respectively intraperitonealy injected with lipopoly saccharide( LPS ,8 0 μg/ kg) to induce fever. The rats of the normal group with physiological saline. Every week we observed every group of changes of autonomous be- havior syndromes. The rectal temperature was measured at different time after ip LPS. And then IL-Iμ,IL-2,GAS,CCK,B-EP,SS in blood were also measured. Result: Compared with the normal group, the rectal temperature of the spleen-qi deficiency with fever group raised signif- icantly at 60min, 120min, 180min( P 〈 0.05 )after ip LPS. The integration of autonomous behavior syndromes increased obviously. The blood of SS, IL-113, IL-2 increased significantly( P 〈 0.05 ). Compared with the spleen-qi deficiency with fever group, the indices of autono- mous behavior syndromes were improved partly by the large dosage of Astragalus mongholicus group. The rectal temperatures which were in different dosage of Astragalus mongholicus group decreased after ip LPS 60min in different degree. Both the temperature of the large dosage of Astragalus group and the medium dosage of Astragalus mongholicus group significantly declined at many different time points after ip LPS( P 〈 0.05). The blood of IL-2 and GAS decreased significantly by the large dosage of Astragalus mongholieus group( P 〈0.05). The blood of SS/CCK obviously decreased or increased by the medium dosage of Astragalus mongholicus group( P 〈0.05). The blood of GAS and SS de- creased significantly by the small dosage of Astragalus mongholicus group( P 〈 0.05 ). Compared with the medium dosage of Astragalus mong- holicus group and the small dosage of Astragalus mongholicus group, the blood of IL-2 decreased significantly by the large dosage of Astraga- lus mongholicus group( P 〈 0.05 ). Conclusion: The different dosages of Astragalus mongholicus group had the regulating effective to the rat model of spleen-qi deficiency with fever in different degree. It was obvious that the large dosage of Astragalus mongholicus group had the sig- nificant effects for the comprehensive indices.
