摘要
实验在完成电生理记录的离体脑片上完成。用电泳法向记录部位导入麦芽凝集素结合的辣根过氧化物酶(WGA-HRP)或荧光金(FG),在和电生理记录相同的条件下孵育脑片8-12h,浸泡固定后做冰冻切片,常规TMB染色,光镜明、暗视野观察,或直接用荧光显微镜观察。在导入示踪剂周围可看到明确的逆行标记细胞和顺行标记纤维与终末。WGA-HRP和FG标记物全电泳中心最远距离,分别达2cm和500μm以上。结果表明,用常规神经解剖学方法可以在离体脑片上显示电生理记录部位神经元局部联系回路。
A method for tracing load ncuronal connectivity in combination with physiological studies on brain slices is described. After electrophysiological studies, a tracer, either wheat germ agglutinin conjugated horseradish peroxidase ( WGA-HRP) or fluoro-gold (FG), was applied iontophorctically to the studied areas on the brain slices. Following incubation for 8-12 hours with artificial cerebrospinal fluid containing 95% O2 and 5% CO2 at 37 ±1℃. A number of labelled neurons and fibers or terminals were found in nearby arcas. The maximum distance of labelling from the application center of the tracer was about 2 an for WGA - HRP and 0.5 mm for FG. The present results show that local neuronal circuits involved in clcctrophysiological studies can be followed on brain slices by tract - tracing techniques.
