摘要
作者用反相高效液相色谱法(岛津LC-6A液相色谱)测定了小鼠血中补骨脂素,分析条件:Shim-Pack CLC-ODS柱,150mm×6mm,用水-甲醇-四氢呋喃(60:25:15)为流动相;定量分析以8-甲氧基补骨脂素为内标,以氯仿一次性提取全血样品,全血中补骨脂索浓度为1,5,10mg/L时,回收率分别为103.7,95.1和98.9%;日内精密度CV分别为3.35,3.90,3.23%;日间精密度CV为7.93,2.92,3.74%。线性范围为0.5-10mg/L时,r为0.9998。小鼠一次ig补骨脂素后属一室模型,其动力学参数:t1/2为3.64h;Ka为4.04 h^(-1);K为0.17h^(-1);C_(max)为3.77mg/L;AUC为26.52mg·h/L。
Whole blood psoralen of mice was detected by reversed - phase high -performance liquid chromatography based on liquid chromatography Shimadzu LC - 6A packed with reversed - phase of Shim - Pack CLC - ODS column ( 150 mm ×6 mm ). The mobile phase consisted of a mixture of water , methanol and tetrahydrofuran (60 : 25 : 15 ) and pumped through the column at the rate of 1.5 ml/min . 8 - Mcthoxypsoralcn (8 - MOP ) was used as an internal standard . Trichlorometnane provided a complete extraction of psoralen and 8 - MOP from the whole blood . The precision of this method was acceptable with the coefficient of variation ranging from 2.92 to 7.39% at the concentration of 1, 5, 10 mg/L, their recovery being 103.7%, 95.1% and 98.9% respectively. The pharmacokinetic characteristics of psoralen were investigated in five healthy mice of each group after oral administration of 50 mg/kg . The results show that the data fit adequately to a one compartment open model with the following parameters : t12 3.64 h , Ka=4.04 h ,1K=0.17 h 1'Cmax= 3.77 mg/ L , AUC=26.52 mg· h/L .
