皖系长毛兔不同周龄Wnt10b、SFRP2基因在皮肤中的表达规律 被引量:9

The Expressions Rules of Wnt10b and SFRP2 Gene in Skin of Wanxi Angora Rabbit

摘要 本研究以被毛长度和被毛密度差异较大的皖系长毛兔(毛用型)和新西兰白兔(肉用型)为实验材料,采用Agilent公司家兔全基因组表达谱芯片筛选差异基因,根据芯片结果,挑出3个与家兔毛纤维生长发育相关的差异基因。实时荧光定量RT-PCR初步验证Wnt10b、Shh、SFRP2基因,检测结果与表达谱芯片结果一致。随后为研究差异表达基因在皖系长毛兔中的表达规律,分别选择一个上调基因(Wnt10b基因)和下调基因(SFRP2基因),采用实时荧光定量RT-PCR法研究Wnt10b、SFRP2基因在皖系长毛兔中的表达规律,进一步验证Wnt10b、SFRP2基因与家兔毛纤维生长发育相关。结果表明:Wnt10b基因在皖系长毛兔毛生长周期中起到一定的调节作用,能够通过诱发毛囊的再生而促进毛的生长发育;SFRP2基因在皖系长毛兔毛生长旺盛期表达量相对较低,在毛生长结束期表达量相对较高,因此SFRP2基因在皖系长毛兔毛生长周期中起到抑制毛生长的作用。 This study chose Wanxi Angora rabbit (wool type rabbit) and New Zealand white rabbit (meat type rabbit) for the test material because of the differences in hair length and hair density We screened differentially expressed genes in two varieties of rabbits' hair density by using Agilent's Rabbit expression profile chip. According to the rusults of gene chip we chose three genes that might be involved in the development process of rabbit hair fiber and affect the rabbit hair length and hair density. WntlOb, Shh and SFRP2 gene were confirmed by real time RT-PCR, the results were the same as the expression profile chip. In order to further validate gene chip results, subsequently, we chose one up-regulated gene (WntlOb) and one down-regulated gene (SFRP2), and then we chose Wanxi Angora Rabbit (hair type) as test material to research the expressions of WntlOb and SFRP2 gene in Wanxi Angora rabbit by using real-time fluorescence quantitative RT-PCR method. The results showed that the expression WntlOb gene played a regulatory role in the the Wanxi Angora rabbit hair growth cycle, it was able to induce the regeneration of hair follicles and promote hair growth; the expression levels of SFRP2 gene were relatively low in Wanxi Angora rabbit hair growth period and they were relatively high at the end of the hair growth period. SFRP2 gene played a role in inhibiting hair growth in the Wanxi Angora rabbit hair growth cycle.
