提高西藏地区马铃薯组培苗移栽成活率的关键技术

Key Techniques for Improving Survival Rate of Transplantation of Potato in virto Plantlets in Tibet

摘要 总结了在西藏高海拔地区昼夜温差大、空气湿度低、光照极强等特殊气候条件下基质移栽脱毒马铃薯组培苗的关键技术。选择河沙与羊粪混合作为基质,严格消毒灭菌,利用MS+0.5mg/L B9培养基培养壮苗,合理炼苗,温室外覆盖遮光率60%以上的遮阳网降低光照,温室内搭建塑料小拱棚提高温度及湿度,温度控制在10一25℃以内,空气湿度保持在85%以上,4-5月移栽。这样,可大大提高温室内组培苗基质移栽的成活率,降低成本,提高生产效率。 Key techniques for transplanting virus-free in vitro plantlets were discussed under special climatic conditions, such as high altitude, large temperature difference between daytime and nighttime, low air humidity and extremely strong sunlight. The mixture of river sand and sheep manure is used as the substrate, which will be sterilized; MS + 0.5 mg/L Daminozide (B9) culture medium is used for the culture of sound seedling and hardening-seedling is properly carried out; the greenhouse is covered by a shading net with the shading coefficient of 60% to reduce the sunlight; a small shed is set up in the greenhouse to increase the temperature and humidity, with the temperature being controlled within 10 - 25℃ and air humidity above 85%; and transplantation is carried out from April to May. All these could greatly improve the survival rate, reduce cost and improve the production efficiency.
