摘要
用 Operon公司 OPA-0 1~ OPJ-2 0 2 0 0个 1 0碱基对随机序列引物对甘蓝型油菜隐性核不育株涪优A和可育株涪优 B、显性核不育株 3 7A和可育株 3 7B、广恢系 DGCR98-3 752、RGCR97-1及 F1 材料基因组DNA进行 RAPD扩增 ,共获得 1 3 3 4条扩增带 ,平均每引物扩增带约 6.7条 ,表明甘蓝型油菜存在遗传多样性。引物 OPA-0 5、OPA-0 9、OPA-1 2、OPA-1 3、OPA-2 0等分别扩增出了大小不同的多态性带 ,初步认为 OPA-1 2 75 0 、OPA-1 2 6 0 0 、OPA-1 2 5 0 0 是与 3 7A不育基因有关的带 ,OPA-0 9330 是与涪优 A不育基因有关的带 ;OPA-1 3 70 0 是与 3 7A不育恢复基因有关的带 ,OPA-2 0 5 0 0 、OPA-2 0 45 0 、OPA-2 0 35 0 是与涪优
Using OPA 01~OPJ 20 200 random 10bp primers from Operon company RAPD amplified 10 genome DNAs of dominant genetic male sterile plant 37A and fertile plant 37B, recessive genetic male sterile plant Fuyou A and fertile plant Fuyou B, poly restored line DGCR98 3752, poly restored line RGCR97 1 and their F 1 hybrid. There were 1334 amplified bands totally and 6.7 bands per primer equally. It indicated that there were abundant genetic polymorphism in Brassica napus L. Primers OPA 05、OPA 09、OPA 12、OPA 13 and OPA 20 amplified several bands of different molecular weight, respectively. OPA 12 750 、OPA 12 600 、OPA 12 500 were the bands related to the dominant genetic male sterile gene of 37A. OPA 09 330 was the band related to the recessive genetic male sterile gene of Fuyou A. OPA 13 700 was the band related to the dominant genetic male fertile gene of 37A. OPA 20 500 、OPA 20 450 and OPA 20 350 were the bands related to the recessive genetic male fertile gene of Fuyou A.
