摘要
生物发光和化学发光在生物医学领域内的应用主要包括细胞学检测、分子生物学、卫生学检测、生物传感器、脂质过氧化检测和药物筛选等六个方面。其中细胞学检测主要是利用细胞内ATP导致的虫荧光素酶生物发光进行活细胞计数 ,目前已实现快速、动态、单细胞分析 ;同时也发现了一些新的与生物或化学发光有关的细胞学指标。分子生物学领域内的应用主要为报告基因和分子杂交 ,近年来又有人推出了生物发光实时DNA测序技术。卫生学检测则主要是利用分子杂交或ATP导致的生物发光检测微生物污染。基于生物及化学发光的生物传感器主要有三类 :酶固化传感器、免疫分子传感器和特异调控因子诱导的报告基因传感器。脂质过氧化检测主要是指利用化学发光检测自由基等活性物质。最近还有人分别推出了基于报告基因和细胞增殖的生物发光药物筛选系统。
Six aspects cell detection, as molecular biology, hygiene inspects, biosensor, lipid peroxides & drug discovery are included in the applications of bioluminescence & chemiluminescence to bio-medical science. The cell detection means cell count through detection of bioluminescence caused by ATP & Luciferase. Now, rapid, continuous, single cell analysis is available. Some other indexes relevant to bioluminescence or chemiluminescence were also found. The applications in molecular biology are mainly report gene & molecular hybridization. A real-time bioluminescent DNA sequencing system was also developed. In hygiene inspects, microbial pollution can be detected through bioluminescence of ATP or molecular hybridization. There are three kinds of biosensors relevant to bioluminescence & chemiluminescence: immobilized enzyme sensor, immunoassay sensor and report gene sensor with specialized regulator. Chemiluminescence can also be applied to detection of free radicals which cause lipid peroxideation. Two drug discovery systems based on report gene & cell proliferation respectively were developed in recent years.
