摘要
目的对低渗条件下竹叶状灰岩内部特有的微生物进行培养,了解竹叶状灰岩内部独特的微生态环境。方法本研究采用低渗稀释培养法、透射电镜、16S rDNA序列分析方法以及培养基改造,对竹叶状灰岩中的微生物进行了初步研究。结果在竹叶状灰岩的培养物中发现了两段未培养细菌16S rDNA的存在;在电镜中观察到两种特殊形态的微生物;采用稀释培养法成功的培养了未培养细菌,且能传代,但经过多种培养基的分离培养,未获得未培养细菌的纯培养。结论竹叶状灰岩内部有特殊形态的未培养细菌存在,本文为进一步研究未培养细菌特有的生存机制和代谢途径奠定了基础。
Objective To study the micro-ecological environment of edgewise conglomerate.Methods The microorganisms in edgewise conglomerate was studied by dilution-culture method,transmission electronmicroscope,16S rDNA sequence analysis and medium reconstitution.Results Two pieces of 16S rDNA sequences from uncultured bacteria were found in the culture of edgewise conglomerate,and two microbes with special morphology were observed under transmission electronmicroscope.The uncultured bacteria were cultured successfully through dilution-culture method and were capable of passage,but pure culture of the uncultured bacteria wasn't gained through isolation and cultivation by several kinds of media.Conclusion Uncultured bacteria with special morphology exist in edgewise conglomerate.This article would provide substantial foundation for the study of the survival mechanisms and metabolic pathway of uncultured bacteria.
