摘要
目的用同位素稀释高效液相-串联质谱(ID-LC-MS/MS)方法检测大鼠肺组织中DNA氧化标志物8-羟基脱氧鸟苷(8-oxo-dGsn)含量,评价长期被动吸烟的大鼠肺组织DNA的氧化损伤程度。方法健康雄性SD(Sprague-Dawleyrats)大鼠20只,随机分为4组:被动吸烟1个月模型组(Smoke-1M组)及其对照组(Control-1M组),被动吸烟4个月模型组(Smoke-4M组)及其对照组(Control-4M组)。被动吸烟组每次烟熏1h,每天2次,每次用烟5支。被动吸烟1个月、4个月后,分别取大鼠肺组织检测其DNA中的8-oxo-dGsn含量。结果被动吸烟1个月后,大鼠肺组织DNA中8-oxo-dGsn含量高于其正常对照组(P<0.05);被动吸烟4个月后,肺组织DNA中8-oxo-dGsn含量比正常对照组增加显著(P<0.01)。结论被动吸烟1个月即可造成大鼠肺组织DNA的氧化损伤,被动吸烟大鼠肺组织中DNA氧化损伤程度随烟龄增长有积累趋势,被动吸烟是肺组织中DNA氧化损伤的重要原因。
Objective To evaluate the oxidative DNA damage in the lungs of male Sprague - Dawley rats exposed to side - stream cigarette smoke, we examined the formation of 8 -oxo- 2' -deoxyguanosine (8 -oxo- dGsn) by isotope dilution HPLC - tandem mass spectrometry (ID - LC - MS/MS). Methods Four groups of rats were used in the exposure protocol. These included rats exposed to smoke for 1 month (Smoke -1M group) and its control (Control- 1M group) , rats exposed to smoke for 4 months (Smoke -4M group) and its corresponding control (Control -4M group). Rats were exposed 2 hours/day and 10 cigarettes/day. The lung tissues were obtained for measuring 8 - oxo - dGsn after exposing for 1 month and 4 months. Results The mean level of 8 - oxo - dGsn in the lung tissues from Smoke - 1 M group was higher than that of control - 1M group ( the difference was statistically significant, P 〈 0. 05) , and after another 3 months, the increase in the number of 8 - oxo - dGsn was more obvious (P 〈 0. 01 ). Conclusion These results suggest that oxidative DNA damage is induced in lung DNA by exposing to side - stream cigarette smoke for only 1 month, and that the chronic exposure (4 months) to cigarette smoke enhance the formation of 8 - oxo - dGsn in rat lungs. This phenomenon of oxidative DNA damage in lung of rats is related to the side - stream cigarette smoke.
