G1Na-TK基因转染联合无环鸟苷(ACV)对人肾癌细胞株GRC-1细胞的体外杀伤作用

摘要 目的 探索 HSV1- TK基因联合应用无环鸟苷(ACV)对人肾癌细胞株 GRC- 1细胞的杀伤作用 .方法 对分别培养及混合培养的 GRC- 1细胞和 GRC- 1/ TK细胞 ,用不同浓度 ACV进行处理 ,然后观察细胞形态及数量的改变 .结果 在 ACV作用下 ,GRC- 1细胞的形态及数量无明显改变 ;GRC- 1/ TK细胞被明显杀伤 ,数量明显减少 ,当含 2 5 .0 %GRC- 1/ TK细胞的混合培养细胞即可观察到明显杀伤作用 .结论  HSV1- TK基因转染联合应用无环鸟苷对人肾癌细胞株 GRC- 1细胞具有明显的杀伤作用 ,并存在较强的“旁观者效应”. AIM To study the killing effect of acyclovir on G1Na TK gene transferred human renal cancer cell line GRC 1 cells in vitro . METHODS GRC 1 cells and GRC 1/TK cells were first cultured separately and together, and then treated with different concentrations of ACV. The morphology of tumor cells was observed and the change of quantity was assayed by trypan blue exclusion. RESULTS The surviving rates of GRC 1/TK cells decreased significantly but no obvious change was observed in the morphology and number of GRC 1 cells exposed to ACV. CONCLUSION Acyclovir has a significant killing effect on G1Na TK gene transferred human renal cancer cell line GRC 1 cells in vitro and there does exist the phenomenon of "bystander effect".
