摘要
运用计算机及实验的方法,在粟酒裂殖酵母(Schizosaccharomyces pombe)中发现和鉴定了一个新的、结构特殊的snoRNA──Z15.该snoRNA由一个独立转录的基因编码,位于酵母2号染色体上Z15长达为92个核苷酸,属于反义snoRNA家族.它除了具有典型的boxC/D结构元素和末端配对区外、还含有分别与酿酒酵母Z15和Z2相同的两个反义序列,反映了粟酒裂殖酵母川Z15sonRNA基因可能通过2种snoRNA基因的重组而来的.推测Z15除了指导25S rRNA A849和A908的2’-氧-核糖的甲基化修饰外,还可能在rRNA构象形成中起作用.粟酒裂殖酵母Z15的发现.从一个新的角度揭示了snoRNA基因结构和功能的复杂性,并为研究这类snoRNA基因的起源和进化提供了重要线索.
A novel small nueleolar RNA, termed Z15, was identified and characterized by the analysis of computer and experiments. Z15 snoRNA, encoded by an independent-transcribed gene on chromosome Ⅱ of S .pombe,is 92nt in length and belongs to the antisense sonRNA family in possessing boxC/D motif and terminal stem-forming sequences. In particularly, the Z15 snoRNA has two antisense sequences identical to that of Saccharomyces cerevisiae Z15 and Z2 snoRNA respectively, this mosaic structure implies that S. pombe Z15 originated probably from two different snoRNAs by gene recombination. According to the structural analysis, S. pombe Z15 is Presumed not only to function as a methylation guide for 25S rRNA at position A849 an fi A908, but also as a RNA chaperon for rRNA conformation.
