摘要
目的:观察丹参酮Ⅱ-A磺酸钠对房颤大鼠心房肌细胞电重构及细胞膜钾离子通道的影响。方法:制备房颤动物模型,荧光定量PCR技术测定心房肌细胞Kv1.5钾通道基因表达,HE染色观察心房肌细胞超微结构变化。结果:丹参酮Ⅱ-A磺酸钠、胺碘酮能明显改善房颤大鼠心房肌细胞钾通道Kv1.5基因表达减低水平(P<0.05)及心房肌细胞组织超微结构。结论:丹参酮Ⅱ-A磺酸钠能够通过改善房颤大鼠钾通道Kv1.5基因表达下调所引发的心房电重构,从而预防和治疗房颤。
Objective: To observed the tanshinone Ⅱ-A sulfonate atrial fibrillation rat atrial myocytes electrical remodeling and cell membrane potassium ion channels. Methods: Preparation of atrial fibrillation animal models, quantitative PCR determination of atrial myocytes Kvl.5 potassium channel gene expression, HE staining atrial myocytes ultrastructural changes. Results: Tanshinone Ⅱ-A sulfonate, amiodarone significantly improved atrial fibrillation rat atrial myocytes potassium channel Kvl.5 gene expression level reduction (P〈0.05) and atrial muscle cell ultrastructure. Conclusion: Tanshinone Ⅱ-A sulfonate by improving atrial fibrillation rats potassium channel Kvl.5 gene expression caused by atrial electrical remodeling cut to the prevention and treatment of atrial fibrillation.
