牦牛、黄牛和绵羊EPO基因部分片段遗传多态性分析 被引量:7

Analysis on genetic polymorphism of the partial fragment of EPO gene in yak、cattle and sheep

摘要 为研究低氧调控促红细胞生成素EPO基因表达的分子机制,本试验选择青藏高原低氧环境中的牦牛和乔科、欧拉、甘加3个藏羊群体及较低海拔的早胜牛和蒙古羊、滩羊、小尾寒羊为研究对象,对EPO基因的部分片段(1 308bp)进行多态性分析。对38个核苷酸序列进行比对分析共得到16个单倍型,84个突变位点,其中有义突变6个。分析发现单倍型Hap_9,10,11,12,13为黄牛特有,Hap_14,15,16为牦牛特有;Hap_1为6个绵羊品种所共有,Hap_3为除小尾寒羊外其他绵羊共有,Hap_6,7为低海拔绵羊小尾寒羊特有,Hap_2为高海拔绵羊甘加羊和乔科羊共有,Hap_5为乔科羊特有,Hap_8为位于中等海拔的蒙古羊特有,Hap_4为乔科与蒙古羊所共有。在该段基因编码的141个氨基酸上,3个物种中共发现6个突变位点。试验结果表明EPO基因多态性丰富,不同海拔物种单倍型分布不同。 In order to research the molecular mechanisms of hypoxic regulation of erythropoietin (EPO) gene expression,the polymorphism of partial fragment (1 308 bp) of the EPO gene in yak, cattle and sheep at different altitudes were analyzed. Through comparing and analyzing the se- quence of the 38 nucleotide, 16 haplotypes and 84 point mutations,including 6 sense mutations, wei'e obtained. The results showed that Hap_9,10,11,12,13 were unique for cattle, Hap_14,15,16 were specific to the yak. Hap_l was unique for six sheep breeds, Hap_3 unique for total sheep breeds except to small tailed Han sheep, Hap_6,7 unique for low-altitude sheep of small tailed Han sheep,Hap_2 unique for the high-altitude breed such as Ganja sheep and Qiaoke sheep,Hap_ 5 were specific to the Qiaoke sheep, Hap 8 unique for the Mongolian sheep in the medium-alti- tude, Hap_4 was owned by Qiaoke and Mongolian sheep. This fragment of the EPO gene encoding 141 amino acids,6 mutations were found in the three breeds. The results of this study showed that EPO gene was polymorphism,haplotype distribution was different at different altitudes animals.
