摘要
目的 探讨反义ＨＳＰ９０降低ＨｅＬａ细胞恶性度的机制。方法 经荧光染色观察细胞形态，ＤＮＡ Ｌａｄｄｅｒ及流式细胞仪检测凋亡细胞。结果 转染有反义ＨＳＰ９０的ＨｅＬａ细胞经荧光染色细胞呈不均一亮蓝色，荧光染色观察可见细胞变小，染色质浓缩并产生凋亡小体，ＤＮＡ Ｌａｄｄｅｒ可见明显的梯形条带，流式细胞仪检测有凋亡的细胞。结论 反义ＨＳＰ９０可诱导细胞凋亡。
Objective To study the mechanisms of HeLa cells inhibited by antisence HSP?90.Methods The apoptosis of HeLa cells were detected with fluorescence stain,DNA Ladder and flow cytometer.Results Fluorescence stain apoptotic bodies are seen after treatment with antisence HSP?90.Observation with fluorescence microscope shows that the cell volume as well as nucleus become smaller,cytoplasm especially chromatin condensed,and the chromatin moves to the inner side of nuclear membrane (margination) and even clots and apoptotic bodies formed.Analysis with flow cytometry shows a sub G1 peak after treatment with antisence HSP?90,indicating programmed cell death happened.Conclusion Antisence HSP?90 induces apoptosis of HeLa cells.
