妊娠糖尿病大鼠视黄醇结合蛋白4水平的变化及与胰岛素抵抗的关系 被引量:2

摘要 目的探讨妊娠糖尿病大鼠视黄醇结合蛋白4(RBP-4)水平的变化及与胰岛素抵抗的关系。方法将20只健康SD孕鼠随机分为妊娠糖尿病大鼠(GDM)组10只、正常妊娠对照组(NC)10只。采用链脲佐菌素(STZ)腹腔一次性注射制备GDM大鼠模型。各组大鼠于分娩前及妊娠20d,测定空腹血糖(FPG)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、空腹胰岛素水平(FINS)和血清RBP-4水平,计算胰岛素抵抗指数(HOMA-IR)。结果①GDM组大鼠血清FPG、FINS、TG、TC、LDL-C、RBP-4、HOMA-IR高于正常妊娠对照组(P<0.05,P<0.01);②Pearson相关分析显示,GDM组大鼠HOMA-IR与FPG、FINS、TG、TC、LDL-C、RBP-4呈正相关(P<0.05,P<0.01)。③多元线性逐步回归分析结果提示,TG、RBP-4是影响GDM组大鼠HOMA-IR独立的危险因素。结论 RBP-4水平的升高与GDM胰岛素抵抗有关。 Objective To observe the serum retinol binding protein 4(RBP-4) change in gestational diabetic rats, and to explore the correlation between RBP-4 and insulin resistance. Methods Twenty Sprague-Dawley rats were randomly divided into 2 groups: GDM group(n= 10)and normal pregnant control group (NC)(n= 10), GDM rat model was established by intraperitoneal injection of streptozotocin(STZ). The rats were sacrificed on the 20-day of pregnancy before delivery. The levels of fasting plasma glucose (FPG), total cholesterol (TC), tri- glycerides (TG), low density lipoprotein cholesterol (LDL-C), fasting insulin (FINS) and RBP-4 were detected. The degree of insulin resistance was measured by insulin resistance index (HOMA-IR). Results The serum levels of FPG, FINS, TG, TC, LDL-C, RBP-4 and HOMA-IR increased significantly in GDM group compared with control group (P〈0.05, P〈0.01). The correlation analysis showed that H OMA-IR was positively correlated with the serum levels of FPG, FINS, TG, TC, LDL-C and RBP-4(P〈0.05, P〈0.01). The multiple stepwise regression analysis indicated that the levels of TG and RBP-4 were independent risk factors for HOMA-IR. Con- clusion The level of serum RBP-4 is significantly higher in GDM rats. RBP-4 is associated with insulin resistance in GDM.
