摘要 目的 观察电化学治疗对人肝癌细胞SMMC772 1化疗耐药性的影响 ,为其临床应用提供实验依据。方法 细胞计数法观察ECT、化疗药阿霉素 (ADR )结合的抑癌效应 ,应用“多药效应分析软件”分析两者之间的相互作用。流式细胞仪 (FCM )分析ECT对细胞内ADR累积的影响 ,免疫组化法检测ECT对SMMC772 1细胞P gp的影响 ,RT PCR法观察ECT对细胞MDR1mRNA水平的影响。 结果 ECT与ADR结合治疗的合并指数 (CI) >1,提示ECT、ADR呈拮抗效应 ,ECT使化疗耐药性增加 ,敏感性下降。ECT 5V1C处理后 2hSMMC772 1细胞内ADR的积累减少 46 .5 % ,ECT 5V2C处理后 2d细胞的MDR1mRNA及P gp表达水平升高。 结论 ECT上调SMMC772 1细胞MDR1基因的表达 ,增加化疗耐药性。 Objective To observe the influence of electrochemical therapy (ECT) on chemoresistance of SMMC7721 cells and to offer the experimental basis for clinical use. Methods The inhibiting effect of ECT combined with ADR on SMMC7721 using cell count method. The“multiple drug effect analysis software ”was used to evaluate the influence between ECT and ADR. The influence of ECT on intracellular ADR accumulation was investigated with flow cytometric (FCM) analysis. Change of MDR1 gene mRNA and protein P-gp after ECT treatment were studied by RT-PCR and immunohistochemistry assay. Results The combined index (CI) of ECT combined with ADR was above one, which suggested that ECT enhanced chemoresistance and reduced chemosensitivity. The intracellular ADR concentration in SMMC7721 decreased by 46.5 % two hours after ECT 5V1C treatment. Expression of MDR1 mRNA and P-gp increased two days after ECT 5V2C treatment. Conclusion ECT enhances the expression of MDR1 gene and the chemoresistance.
