摘要
为获得更高含量的可溶性N蛋白,对构建的重组大肠杆菌E.coli BL21(DE3)(pET-32a-N)的表达条件(如诱导温度、诱导时间、IPTG浓度)进行了优化。结果表明:N蛋白最佳诱导表达条件为18℃诱导17 h、IPTG浓度0.2 mmol/L;优化后可溶性N蛋白的表达量为22.1 mg/L,比优化前37℃、IPTG浓度1.0 mmol/L条件下诱导4 h提高了58.5%。
In order to gain a higher content of soluble N protein, the expression conditions of recombinant E. coli BL21 (DE3)(pET-32a-N), such as induction temperature, induction time and IPTG concentration, were optimized by using polyacrylamide gel electrophoresis (SDS-PAGE)and electropherogram analyzing software. The results showed that the optimal expression conditions were an induction at 18 ℃ and 0.2 mmol/L IPTG for 17 h. Under the optimal conditions the concentration of soluble N protein was 22.1 mg/L, increasing by 55.8% over that under induction at 37 ℃ and 1.0 mmol/L IPTG for 4 h.
