肾母细胞瘤微卫星DNA变化分析

摘要 目的 研究分析 13例肾母细胞瘤 (Wilms肿瘤 )患儿 5个染色体位点微卫星标记的变化。方法  5个微卫星标记分别为 :D7S180 8、D9S16 1、D12S10 5 6、D17S791和D2 2S2 74,应用PCR方法扩增Wilms肿瘤组织DNA ,以该患儿正常组织作对照 ,扩增产物用含脲素的聚丙烯酰胺凝胶电泳及银染技术进行分析。结果  13例Wilms肿瘤患儿检测 5条染色体 5个位点的微卫星多态标记 ,只1例患儿 7号染色体D7S180 8位点发现存在不稳定 ,在其他 4个位点均未发现有微卫星标记的变化。结论 胚胎恶性肿瘤发生机制可能与成人肿瘤不同 ,微卫星DNA变化在Wilms肿瘤不常见。染色体 7p可能存在与Wilms肿瘤发生相关基因。 Objective Microsatellite DNA are short tandem repeat sequences and are useful DNA markers distributed throughout the genome. We have investigated the microsatellite instabilities (MIN) of five chromosome loci in 13 Wilms' tumors. Methods Five microsatellite DNA markers including D7S1808, D9S161, D12S1056, D17S791 and D22S274 from the Wilms' tumor specimens and normal tissue of the same patients were amplified with PCR. The sequences were analysed on polyacrylamide gel containing urea and silver. Results Somatic instability at microsatellite repeat sequence D7S1808 was found in 1 out of 13 Wilms' tumor specimens. No alteration was detected in the other four loci. Conclusions Microsatellite instability, unlike that of adult tumors, is rare in Wilms' tumor. The gene implicated in Wilms' tumor may be located in chromosome 7p.
