摘要
筛选油橄榄叶不同提取部位中清除自由基最佳活性部位。油橄榄叶经乙醇回流提取、大孔吸附树脂分离富集,得到油橄榄叶不同提取部位A-F。分别采用DPPH.法和ABTS+.法对油橄榄叶不同提取部位进行清除自由基活性评价,并用高效液相色谱(HPLC)法探讨最佳活性部位的物质基础。结果表明,油橄榄叶不同提取部位均有清除自由基活性。其中,对于DPPH.的清除能力依次为C>D>E=F>A>B,对于ABTS+.的清除能力依次为C>D=F>E>A>B。通过对部位C进行HPLC分析,并用对照品标定色谱图中色谱峰,最高峰为橄榄苦苷。说明部位C为清除自由基活性最佳活性部位,且橄榄苦苷是其主要的活性物质之一。
Screen on the most active fraction of scavenging free radical in the different extract fractions from Olea europaea L. leaves. Different fractions A-F were obtained using reflux by ethanol and separation by macroporous adsorption resin from Olea europaea L. leaves. DPPH. and ABTS+" assays were employed to evaluate the active fractions of scavenging free radical of the different fractions from Olea europaea L. leaves, respectively. The active component in the most active fractions of scavenging free radical was traced by HPLC. The results show that all extract fracttions displayed the different free radical scavenging activity. The seriatim of activity of scavenging DPPH. was C〉D〉E=F〉A〉B and that of scavenging ABTS+. was C〉D=F〉E〉A〉B, successively. Through the HPLC chromatogram of fraction C with that of the reference standard analyzed, it was found that the highest chromatographic peak is oleuropein. Fraction C posses the most activity of scavenging flee radical, and oleuropein is one of the main active components.
