摘要
图位克隆技术属于正向遗传学的范畴,它以突变表型在遗传群体中的分离为出发点,致力寻找导致突变的遗传基础,最终克隆出目的基因。本文简要介绍了图位克隆技术的遗传定位原理以及在玉米基因分离中的关键技术环节,包括遗传群体的构建,多态性分子标记的筛选,目的基因的初定位,精细定位,候选基因的获得、基因的功能验证等,最后分析了图位克隆技术在玉米中应用可能遇到的问题以及可采取的解决措施。
Map-based cloning technology, as one of powerful tools in the forward genetics, was being used to identify the plant mutant genes based on the mutant phenotype segregation in the genetic population. In this review, we briefly introduced the principle of the map-based cloning technology and the key steps in the application of gene isolation in maize, including construction of genetic population, polymorphism screening of molecular markers, gene rough mapping, gene fine mapping, and candidate gene confirmation, and gene function confirmation, and finally discussed the possible problems and solutions in the applications.
