摘要
目的通过供鼠淋巴细胞输注诱导小鼠慢性GVHD,并在此基础上荷瘤建立慢性GVHD的GVL模型,为研究慢性GVHD和GVL作用奠定基础。方法以雄性BALB/c(H-2d)为供鼠,雌性CB6F1(BALB/c×C57BL/6 F1,H-2d/b)为受鼠,通过尾静脉输注8×107/个脾细胞诱导小鼠慢性移植物抗宿主病,在输注25天后,受鼠随机分组,通过尾静脉或腹腔或皮下方式接种P815肿瘤细胞,构建慢性GVHD荷瘤模型,用于研究慢性GVHD的同时研究GVT作用。结果 3种荷瘤方式均可形成荷瘤。尾静脉方式可形成小鼠白血病模型,符合人类疾病特点,成功构建慢性GVDH荷瘤的GVL模型。结论成功建立小鼠慢性移植物抗宿主病荷瘤模型。
Objective Induce murine model of chronic GVHD after donor lymphocyte infusion. In order to study GVL effect at the same time inoculation of tumor cells after transplantation. Methods Male BALB/c( H - 2d) mice were used as donors and female CB6F1 (BALB/c × C57BL/6 F1 ,H- 2d/b) mice as recipients. 8 x 107of spleen cells by tail vein injection to induce murine model of chronic graft- versus- host disease,in experiments meant to study graft- versus- leukemia (GVL) activity, select groups of CB6F1 recipients were injected intraperitoneally (i. p. ) or subcutaneous or intravenous (i. v. ) 25 days after transplantation with P815 tumor cells. Results These three methods all can established the P815 tumor- bearing mice model. Tail vein approach is more in line with characteristics of human disease,the GVL model was established successfully by intraparitoneal injection. Conclusion successfully established of cGVHD mice bearing leukemia model.
