摘要
目的 :探索经尿道给予前列腺素E1治疗勃起功能障碍的可行性、安全性及剂量与效果的关系。 方法 :采用单盲、安慰剂及自身逐级增量给药 (15 0～ 12 0 0 μg)对照观察 2 4例勃起障碍病人的剂量与效果 (阴茎勃起角度、周径、长度变化及持续时间 )的关系。 结果 :发现 6 0 0 μg以内剂量与反应呈直线正相关 ,随剂量增大而阴茎勃起质量增加 ,约 80 %以上病人可达最佳效果 ;6 0 0 μg以上仅有 2 0 %以下的病人有进一步反应 ,且副作用增多。 结论 :PGE1尿道给药疗效确实、安全可靠 ,单独使用剂量应控制在 6 0 0 μg以下。
WT5”HX]Objectives: [WT5”BZ]To evaluate the feasibility, safety and correlation between the dosage and effectiveness of prostaglandin E 1(PGE 1) with medicated urethral system for erection (MUSE). [WT5”HX]Methods:[WT5”BZ] A single blind, placebo controlled study of increasing dosages (150～ 1 200μg ) was performed. 24 men with erection dysfunction were monitored in real time for 2 hours after MUSE, the penial erecting angle (PEA), tumescence and duration of erection were measured. [WT5”HX]Results:[WT5”BZ] The initial response to escalating dosages of PGE 1 from 150μg to 600μg demonstrated a steep dosage dependent increase; at greater than 600μg indicating a nonliner response. More than 80% of the patients attained the maximal response at dosage of 600μg or less and less than 20% benefited from a further increase. [WT5”HX]Conclusions: [WT5”BZ]The effects of PGE 1 with MUSE appear to be receptor dependent and PGE 1 monotherapy for erection dysfunction could be limiled to 600μg or less, since larger amounts offer little additional benefit.
