摘要
为验证中药乳宁Ⅱ号对乳腺癌的治疗作用，本文选用人乳腺癌细胞株ＭＣＦ－７作为研究对象，观察乳 宁Ⅱ号对其体内外生长的影响。结果表明，含中药乳宁Ⅱ号的药物血清在培养液中浓度为２０％和３０％时对 ＭＣＦ－７的体外生长的抑制率达２４．４％、２４．２％（Ｐ＜０．０５），流式细胞仪检测结果说明其机理可能是抑制癌细胞 的ＤＮＡ合成，阻滞癌细胞于Ｇｏ／Ｇ１期，减少Ｓ期比率；同时用中药乳宁Ⅱ号灌饲ＭＣＦ－７荷瘤裸小鼠的结果显 示，乳宁Ⅱ号在剂量为３６ｇ／ｋｇ、１８ｇ／ｋｇ、９ｇ／ｋｇ时抑癌率分别为４０、９％、ｓ５１、４１．３％（Ｐ＜０．０１），并对其病理变化 有一定影响。说明中药乳宁Ⅱ号对人乳腺癌ＭＣＦ－７的生长确有抑制作用。
Using cellculture, serologic pharmacological method and treatment of nude mice bearing established MCF-7turmors with Runing No.2, we ob- served the effects of Chinese herbal medicine Runing NO.2 on MCF-7 human breast cancer cell lines in vivo and in vitro. Invitro test results showed that exposure to rat serum containing Runing No. 2(serum concentration 20%, 30%) resulted in depression of growth of MCF-7 with depression rate 24.2%.respectively. Subsequent cell cycle analysis, using DAN flow cytometry, showed that chinese herbal medicine probable block the systhethize of DNA, retaineted cells in G0-G1 . Invivo rest result indicated that Runing No.2 had inhibition effect on tumor with tumor regression rate 40.9%,51%,41.3% respctively, with regard to dose 36g/kg,18g/kg,9g/kg.
