低剂量γ射线与香烟对胎鼠脑蛋白质及热休克蛋白的影响

摘要 目的 探讨低剂量γ射线、香烟及其联合作用对脑蛋白质及热休克蛋白 (HSP70 )合成的影响。方法 利用Westerndotblot和Lowrey方法对经低剂量γ射线 (6 0 Co,0~ 1.0Gy)和香烟烟雾 (0~ 2支 /只 )处理的孕鼠的胎脑蛋白质和HSP70进行定量研究。结果  0 .2 5Gy以上的照射和高浓度的香烟烟雾(2支 /只 )与对照组比较胎脑蛋白质明显减少 ,并且低剂量γ射线与香烟具有联合作用 ;低剂量γ射线、香烟烟雾及其联合作用下 ,胎脑HSP70明显升高 (P <0 .0 1) ,其中高照射组和高浓度香烟组脑HSP70升高明显低于低照射组和低浓度香烟组 (P <0 .0 1) ,照射与高浓度的香烟组联合明显较单独照射组低 ,有剂量依存关系。结论 低剂量γ射线、香烟及其联合可引起脑蛋白质的含量减少 ,并诱导胎脑HSP70的合成 ,提示胎脑发育损伤与脑蛋白和HSP70的合成有一定关系。 Objective To explore the effect of low dose γ-radiation or cigarette smoking and their combined on the synthesis of protein and heat stress protein(HSP70) in embryonic brains of mice. Methods Protein and HSP70 in embryonic brains of mice were determined with Lowrey method and Western dot bloting following exposure in low dose γ-radiation( 60 Co,0~1.0Gy) and cigarette smoking (0~2.0 cigarette / mouse) . Results Protein contents in embryonic brains of mice in the 0.5 Gy group,1.0Gy group and high concentration cigarette smoking group(2 cigarette/ mouse) were lower than those of the control. The significant synergistic action of low dose γ-radiation and cigarette smoking was found . The HSP70 increased significantly in embryonic brains of mice exposed to low dose γ-radiation and cigarette smoking , and the increase of HSP70 in embryonic brains with high dose γ-radition group and high concentration cigarette smoking group was less than low dose γ-radiation group and low concentration cigarette smoking group , and embryonic brains HPS70 in the mice of combined effect group was decreased compared to the single γ-radiation group . There was dose dependent relationship between γ-radiation and cigarette smoking group.Conclusion The low dose γ-radiation or cigarette smoking and their combined may decrease protein content of embryonic brain and induce the synthesis of HSP70 . The results suggest that there are correlation between the damage of embryonic brain and the synthesis of the protein and HSP70.
