亚硫酸氢钠穿心莲内脂对小鼠肾毒性及其蛋白表达分析

摘要 目的:观察亚硫酸氢钠穿心莲内脂(ASB)的肾毒性机制。方法:20只雄性ICR小鼠随机分为正常对照组和ASB组(1 000mg/kg),每组10只。小鼠尾静脉注射ASB,每天1次,连续7d,正常对照组用等体积0.9%氯化钠溶液同条件予以注射。进行血常规、血液生化指标及肾病理组织学检查。另取一肾提取蛋白质进行双向凝胶电泳分离找出差异蛋白点。结果:ASB组,外周血白细胞从(10.50±2.00)×109/L升高至(13.70±3.40)×109/L,血小板从(666.05±122.09)×l09/L降低至(451.31±207.22)×109/L,淋巴细胞比例从(0.83±0.03)降低至(0.68±0.11);血清尿素氮、肌酐异常升高(P<0.05);其中8只小鼠出现肾脏病变,多为肾小管间质炎症细胞浸润(间质性肾炎)。肾组织蛋白经双向电泳后,ASB组与正常对照组比较有8个差异蛋白点:2个表达增强,2个表达降低,新出现2个,消失1个,移位1个,有显著性差异变化(P<0.05)。结论:在通过肾组织蛋白表达分析后,ASB所致小鼠急性肾损伤且组织有明显不同蛋白差异点表达,这些差异蛋白质可能参与了ASB肾毒性损伤的过程。 Objective: To study the mechanism of nephrotoxicity by andrographolide sodium bisulfite (ASB). Methods: 20 male ICR mice were randomly divided into normal control group and ASB1 000mg/kg group. Mice in ASB group were given ASB 1 000mg/kg by tail vein, once daily for 7d. The normal control group was given an equal volume of sailine at the same conditions. Examining blood routine and blood biochemical indexes detection; taking a kidney for histopathological examination. Another kidney were homogenated and renal proteins were extracted, differential proteins were expressed by the two-dimensional (2-D) electrophoresis proteomic. Results: In ASB 1 000mg/kg, white blood cells elevated from(10.50±2.00)x109/L to (13.70±3.40)x109/L. Platelets reduced from(666.05±122.09)x109/L to (451.31±207.22)x109/L, the proportion of lymphocytes reduced from(0.83±0.03) to (0.68±0.11); serum BUN, CR were elevated respectively (P〈0.05); 8/10 mice in this group had renal lesions which mostly were renal tubulointerstitial inflammatory cells infiltration(interstitial nephritis). 8 protein spots were obtained after two-dimensional electrophoresis: there were 2 spots expression increasing, 2 spots expression decreasing, 2 new protein spots emerging, 1 spot disappearing, 1 spot shift in treatment group. There were some significant changes (P〈0.05). Conclusion: Different spots were expressed between the two groups, and different protein might be participated in the course of injury kidney by ASB.
