摘要
目的:观察甲基强的松龙对局灶性脑缺血模型纹状体内TNFα、NO含量的影响。方法:用线栓法制备局灶性脑缺血模型,用ELISA方法检测TNFα,采用Griess试剂测定微透析液中NO的代谢产物亚硝酸盐含量。结果:缺血1h再灌注6h,缺血侧纹状体内NO及TNFα含量升高,给予3mg·kg-1甲基强的松龙,可以使两种物质的含量下降。结论:给予小剂量的甲基强的松龙(3mg·kg-1)可以降低脑缺血组织的TNFα、NO含量,可能对脑缺血损伤产生保护作用。
Aim: Effects of methylprednisolone on the level of TNFα, NO in the ischemic striatum from focal ischemic- reperfusion rats were observed. Methods: The focal cerebral ischemia model was made by occluding the middle cerebral artery with an intraluminal suture. TNFα was measured with ELISA Kits. The NO formation in the microdialysis fluids was evaluat- ed based on Griess reaction. Results :The levels of TNFα and NO increased significantly after 6 h reperfusion followed by 1 h ischemia. The levels of TNFα and NO in the ischemic striatum decreased compared with normal saline group after given 3 mg/ kg methylprednisolone. Conclusion: Lower dose of methylprednisolone (3 mg/kg)reduced TNFα and NO production in the is- chemic striatum. It may play a neuroprotective role for cerebral ischemia.
