摘要
目的总结一步法抽提人体肝细胞癌(HCC)癌组织及实验性肝纤维化肝组织总RNA的体会。方法参照Chomczynski一步法抽提12例人体HCC癌组织及16例大鼠纤维化肝组织总RNA。结果及体会 28例标本一次性抽提成功,且得率较高。本研究体会到,保持标本新鲜,少坏死,避免内源性RNA酶(RNase)活化及外源性RNase污染是一次性抽提成功的关键。
Objective To summarize the experience in extraction of total RNA from the tissues of human hepatocellular carcinoma (HCC) and experimental hepatic fibrosis (EHF). Methods The total RNA in the tissues from 12 patients with HCC and 16 rats with EHF was extracted with Chomczynski's one-step method with slight modification. Results The total RNA was successfully extracted. Conclusion Keeping the tissues fresh and avoiding activation and contamination of RNase is crucial in tissue RNA extraction.
