大鼠严重烫伤淋巴组织细胞凋亡的原位TUNEL及电镜观察 被引量:1

摘要 目的 :观察严重烧伤后大鼠淋巴组织内细胞凋亡状况。方法 :采用大鼠 30 %体表面积Ⅲ度烫伤模型 ,应用原位组织TUNEL技术及透射电镜 ,分别观察烫伤后 3、6、12h 3个时相点大鼠胸腺、脾、肠系膜淋巴结凋亡细胞的分布、定位和形态特征。结果 :烫伤后 6、12h ,在胸腺、脾、肠系膜淋巴结出现明显的细胞凋亡 ,并见其呈散在分布于胸腺的皮质、脾的白髓及肠系膜淋巴结的皮质淋巴小结之间。结论 :严重烫伤可致大鼠淋巴组织的细胞凋亡。组织原位观察结果表明细胞凋亡主要发生于T细胞聚集区 。 Purpose To explore the histological features of apoptotic cell death of lymphoid tissue and the relationship between lymphocyte apoptosis and postburn immunosuppression in severely burned rats. Methods Thirty five SD rats were divided randomly into two groups: burn group ( n =30) and control group ( n =5). The rats of the burn group then sustained a 30% TBSA Ⅲ degree scald injury and the control group underwent a sham scald procedure. Lymphocyte apoptosis of thymus ,spleen and mesentery lymph node was detected by using in situ terminal deoxynucleotidyl transferase mediated Dig dUTP nick end labeling (TUNEL)technique and electron microscopy. Tissue specimens were taken at 3,6 and 12 hours after scald or sham scald. Results At 6, 12 hours after thermal injury, It was detected that the number of lymphocyte apoptosis was increased significantly in thymus, spleen and mesentery lymph nodes. The apoptotic lymphocytes mainly disseminated at T cell residing area. Conclusion The result indicated that lymphocytes apoptosis of thymus, spleen and mesentery lymph nodes might be induced by thermal injury at early stage after burn injury and they might mainly belong to the T cell family. These novel observations suggested that apoptosis may play a major role in thermal injury related cellular immunodeficiency.
