摘要
目的 :从人胎盘组织基因组 DNA中 ,克隆人神经营养因子 - 4/ 5 (h NT- 4/ 5 )基因。方法 :经 PCR扩增出的 DNA片段用 Pst I单酶切后 ,酶切图谱与文献报道一致。再将大量扩增后的目的 DNA片段连接于测序载体M13m p18,用手工测序和 ABI373A DNA自动序列分析仪自动测序。结果 :两项分析结果都确证所得到的目的基因与天然 h NT- 4/ 5编码序列一致。 结论 :首次成功地从人胎盘组织克隆出人神经营养因子 - 4/
Objectives: With polymerize chain reaction (PCR), human neurotrophin 4/5 gene (hNT 4/5) was successfully cloned from the genome of human placenta histiocyte by our study. Methods: Using the Pst I, the result accorded with the documents. After PCR, the DNA joined the sequencing carrier M13 mp18. Results: Manual and auto analyzing instrument (ABI 373A) sequence were all confirmed that the DNA was hNT 4/5. Conclusion: With PCR, hNT 4/5 was successfully cloned from human placenta.
