摘要
探讨人急性髓细胞性白血病 (acutemyeloidleukemia,AML)细胞mdm2及p53基因蛋白表达水平、相互关系及与临床预后的关系。方法 :用免疫组化方法测定mdm2及 p53蛋白表达 ,HPIAS 1000高清晰度病理图像细胞测定系统分析结果。结果 :75%的AML患者表达不同程度的mdm2蛋白 ,且在各种分型中均有mdm2蛋白过度表达者 ;18 75%的AML患者表达p53蛋白;在AML患者中mdm2蛋白阳性同时p53蛋白阴性者占65 63 % ;mdm2表达阳性者其完全缓解率 (29 17% )较mdm2阴性者 (83 3 %)明显减低(P<0 01),mdm2与 p53蛋白均阳性者缓解率最低,而两种蛋白均阴性者均获完全缓解。结论 :在AML患者中有较高水平的mdm2蛋白表达,以mdm2阳性( +)、p53阴性( -)为其主要表达方式 。
Objective: To explore mdm2 and p53 gene expressions in human acute myeloid leukemia(AML) cells, relationship between each other and their predictive value of chemotherapeutic efficacy Methods: The mdm2 and p53 gene expressions were determined by immunohistochemical staining and analysed by using high resolution pathological image analysis system Results:75%of AML patients was detected mdm2 protein in virous degree,which occured in any subtype leukemia The expression rate of p53 protein was 18 75%in 32 AML patients Furthermore,mdm2 (+)but p53 (-)was accounted for 65 63% The complete remission (CR)rate in mdm2 (+) patients (29 17%) was significantly lower than that in mdm2 (-) patients (83 3%)(P<0 01) The patients had lowest CR rate if their AML cells were both positive for mdm2 and p53 expressions Conclusions:Most of AML patients express mdm2 gene and,mdm2 (+),p53 (-) is the main expression pattern in AML patients Different expression patterns of the two proteins are related to the clinical outcome
