人羊膜上皮细胞治疗创伤性脊髓损伤的移植免疫研究 被引量:1

The transplantation immunity study of human amniotic epithelial cells transplantation on treating spinal cord injuries

摘要 目的探讨人羊膜上皮细胞移植治疗大鼠创伤性脊髓损伤的作用及细胞移植后免疫反应情况。方法采取大鼠脊髓半横断方法建立动物模型后损伤局部直接注射移植细胞,设三组:羊膜上皮细胞移植组、骨髓源间充质细胞移植组、PBS液对照组;利用BassoBeattieBresnahan运动功能评分(BBB评分)评价运动功能恢复情况,用ELISA法测定大鼠血清IL-2和IL-10水平、免疫组织化学法标记CD3+T细胞浸润情况评价移植免疫情况。结果(1)BBB评分显示,三组大鼠在移植后3周内均有较快恢复并在3周后进入平台期,细胞移植组恢复更明显且羊膜上皮细胞组优于骨髓源问充质细胞组(P〈0.05)。(2)羊膜上皮细胞组大鼠血清IL-2的水平较骨髓源间充质细胞组明显偏低,IL-10水平则偏高。(3)CD3免疫组化显示各组切片在早期均有CD3+细胞浸润,移植后3个月羊膜上皮细胞组CD3+细胞浸润明显少于骨髓源间充质细胞组(P〈0.05)。结论细胞移植治疗创伤性脊髓损伤效果明显;羊膜上皮细胞免疫原性较骨髓源间充质细胞弱。 Objective To discuss the role of human amniotic epithelial cells (HAECs) transplantation in the treatment of traumatic spinal cord injured rats and cell transplantation immune reaction conditions. Methods Take rat spinal cord half transection method to establish animal models, inject transplantation cells directly into damaged local, and set up three groups for comparison: HAECs transplantation group, bone marrow stromal cells (BMSCs) transplantation group, and PBS liquid group. Assessed movement function recovery with the Basso - Beattie - Bresnahan movement function score ( BBB scores), evaluated transplantation immunity situation through the determination of rat serum IL - 2 and IL- 10 levels via the ELISA method, and CD3+T cell infiltration with immunohistochemistry. Results (1)BBB scores show that all three groups recovered rapidly in 3 weeks after transplantation and into the platform period after 3 weeks, cell groups' scores are higher and HAECs were higher than BMSCs ( P 〈 0. 05). (2)The level of IL -2 was obviously less in the transplantation group than those in the BMSCs transplantation group ( P 〈 0. 05 ). Meanwhile, IL- 10 was contrary to the IL- 2. (3)The quantity of CD3 + cells in all groups was much at the acute period, but the quantity of CD3+ cells in BMSCs transplantation group is more than HAECs at the chronic period 3 months after cell transplantation ( P 〈 0. 05 ). Conclusions Compared with the PBS group, cell groups play an important value in the treatment of SCI and compared with BMSCs, the HAECs have much Lower immunogenicity.
