摘要
目的 :观察成年小鼠和老年小鼠腹腔巨噬细胞HSP7O .iNOS及iNOSmRNA表达的情况 ,探讨成年和老年机体免疫功能差异的机制 ,为老年医疗保健提供理论依据。方法 :取成年小鼠和老年小鼠各 10只 ,取小鼠腹腔巨噬细胞制作标本 ,采用免疫组化、组织化学、原位杂交及完整细胞斑点印迹技术检测HSP70、iNOS及iNOSm RNA的表达 ,并对所有斑点印迹信号进行扫描 ,其扫描值采用t检验进行统计学处理。结果 :成年小鼠HS7O .i NOSmRNA的表达为 7.13± 2 .12、5.57± 1.36及 12 .39± 2 .6 8,老年小鼠HSP70 .iNOS及iNOSmRNA的表达为 4 .34± 1.57、3.0 3± 1.2 8及 5.89± 1.90 ,成年小鼠和老年小鼠相比有显著差异 ,P <0 .0 1。结论 :提示老年机体免疫功能低下的机制之一可能与机体表达HSP70 .iNOS及iNOSmRNA的下降有关。
Objective:To observe the adult and aged rats abdominal cavity macrophage expression of HSP70,iNOS and iNOSmRNA,and to study the adult and aged body's mechanism of the difference immune function in order to provide theory basis for the aged people's health care .Methods:Samples of rat's abdominal macrophage were taken respectively from 10 adult and aged rats.Immunohistochemistry,tissue chemistry, hybridization in situ and complete cells dot blob technique were used to check expression of HSP70,iNOS and iNOSmRNA,and all dot blob signal were scanned the scanning date of which were statistically handled,Result:Adult rats expression of HSP70 ,iNOS and iNOSmRNA are 7.13±2.12、 5.57±1.36 and 12.39±2.68,while the aged rats expression of HSP70、iNOS and iNOSmRNA are 4.34±1.57、3.03±1.28 and 5.89±1.90. The adult's expression compared with aged rat's, there is great difference in expression of HSP70、iNOS and iNOSmRNA.Conclusion:It is suggested that the aged body's mechanism of immune function may be related to aged body's lower expression of HSP70,iNOS and iNOSmRNA.
