人胚胎干细胞向间充质干细胞分化的初步研究

摘要 目的建立人胚胎干细胞(hESCs)向间充质干细胞(MSCs)分化方法,并初步鉴定hESCs来源的MSCs(hESC-MSCs)。方法用拟胚体法将hESCs分化为MSCs,对其形态进行观察;用流式细胞术检测细胞表面标志;测定细胞化学染色特性。结果 hESC-MSCs形态与人骨髓间充质干细胞(hBM-MSCs)相似;流式细胞分析显示hESC-MSCs表达CD29、CD44、CD13,但不表达CD14、CD133、CD34、CD45、HLA-DR;细胞化学染色显示,hESC-MSCs碱性磷酸酶阴性,过碘酸-雪夫反应弱阳性。结论hESCs在体外可分化为hESC-MSCs,hESC-MSCs符合MSCs特性。 Abstract: Objective To establish a method that human embryonic stem cells (hESCs) were differentiated into mesenchymal stem cells (MSCs) and identify the human embryonic stem cell-derived mesenchymal stem ceils (hESC-MSCs). Methods hESCs were differentiated into MSCs by the method of embryoid body formation. The morphology and cytochemical staining of hESC-MSCs were ob- served, and the surface antigens determined by flow cytometry. Results The morphology of hESC-MSCs was similar to that of human bone marrow mesenchymal stem cells (hBM-MSCs). Flow cytometry showed that CD13, CIM4 and CD29, but not CD14, CD133, CD34, CIM5 and HLA-DR, were expressed in hESC-MSCs. The cytochemical staining showed that there was negative ALP and weakly positive periodic acid-Schiff (PAS) reaction in hESC-MSCs. Conclusion hESCs could be differentiated into MSCs in vtro, and the characteristics of hESC-MSCs was similar to that of MSCs.
