摘要
目的研究牙龈卟啉单胞菌(P.gingivalis)上清液对兔血管平滑肌细胞增殖和迁移的影响.方法组织块法进行兔腹主动脉VSMCs的原代培养,复苏培养P.gingivalis标准菌株,不同浓度细菌上清液刺激细胞48h,MTT法检测细菌上清液对细胞增殖的影响;爬片法检测7 d内4.3×106CFU/mL的P.gingivalis上清对细胞迁移的影响.结果浓度高于4.3×106CFU/mL的P.gingivalis上清液能抑制VSMCs的增殖;4.3×106CFU/mLP.gingi-valis上清作用于VSMCs前4 d能促进VSMCs的迁移,明显高于对照组(P<0.05).结论高于4.3×106CFU/mL的P.gingivalis上清液能抑制VSMCs的增殖,4.3×106CFU/mL P.gingivalis上清液可促进血管平滑肌早期的迁移,从而在动脉粥样硬化的发生发展过程中可能发挥一定的作用.
Objective To explore the effects of proliferation and migration on the rabbit vascular smooth muscle cells stimulated by the supernatant of Porphyromonas gingivalis.Methods Tissue block method was used to primary cluture the VSMCs derived from rabbit abdominal aorta.The standard P.gingivalis lines was resuscitated and cultured.The MTT was employed to assay the proliferation of VSMCs stimulated for 48h by the supernatant.Measured the distances of VSMCs by climbing film law in 7 days after the treatment of the 4.3×106CFU/mL P.gingivalis supernatant.Results The concentrations higher than 4.3×106 CFU/ml P.gingivalis supernatant could obviously inhibited the proliferation of VSMCs.And after the treatment of 4.3×106 CFU/mL P.gingivalis supernatant in 4 days,the migration distance of VSMCs was significant higher than the control group(P0.05).Conclusions The concentrations higher than 4.3×106CFU/mL P.gingivalis supernatant could inhibite the priliferation of VSMCs.4.3×106CFU/mL P.gingivalis supernatant could improve its early migration.So the P.gingivalis may play the role in the progress of the generation and development of the AS.
