β-榄香烯联合紫杉醇诱导胃癌细胞凋亡的体外研究 被引量:12

β-elemene combined with paclitaxel induced cell apoptosis in human gastric cancer cells

摘要 目的:观察联合β-榄香烯和紫杉醇对诱导胃癌细胞凋亡以及对凋亡相关蛋白表达的影响。方法:β-榄香烯和紫杉醇单独及联合作用于胃癌SGC7901细胞,MTT法检测细胞增殖抑制率,流式细胞术检测细胞凋亡,免疫印记检测凋亡相关蛋白的表达。结果:单药β-榄香烯作用SGC7901细胞24、48和72小时的IC50值分别为(52.56±6.26)、(28.69±2.36)和(13.33±0.64)μg/ml。单药紫杉醇作用24、48和72小时的IC50值为(7.23±1.98)、(1.72±0.76)和(0.22±0.19)μg/ml。选用低浓度药物(10μg/ml的β-榄香烯和2μg/ml的紫杉醇)联合处理SGC7901细胞24小时,与相应浓度β-榄香烯、紫杉醇单药组相比,两药联合组对SGC7901的增殖抑制率显著增强(P<0.05),细胞凋亡率显著提高。进一步研究发现β-榄香烯和紫杉醇联合后,诱导了PARP裂解以及显著下调了Bcl-2与Bax的比值。结论:β-榄香烯和紫杉醇联合用药后,可能通过下调Bcl-2与Bax的比值诱导PARP裂解,进而抑制胃癌细胞增殖和诱导细胞凋亡。 Objective: To explore the cooperative effect of β - elemene and paclitaxel on cell apoptosis and apoptosis -related protein expression in human gastric cancer SGC7901 cells. Methods:SGC7901 cells were treated withβ-elemene and/or paclitaxel. Cell proliferation was assessed by MTT assay. Cell apoptosis was determined rising fl0w eytometry. Protein expression was detected by western blot. Results: β - elemene ( 10μg/ml) and paclitaxel (2μg/ ml) synergistically inhibited cell proliferation in SGC7901 cells. Flow eytometry showed that the combination of 15 - elemene and paelitaxel significantly induced cell apoptosis, the clearage of PARP, and the ratio of Bcl - 2 and Bax. Conclusion: β- elemene and paclitaxel promoted cell apoptosis and regulated apoptosis -related protein expression in human gastric cancer SGC7901 cells.
