蜕皮甾酮对大鼠蛛网膜下隙出血后大脑皮质损伤的影响

摘要 目的探讨蜕皮甾酮对大鼠蛛网膜下隙出血后大脑皮质损伤的保护作用及其机制。方法 60只雄性SD大鼠,随机分为假手术组、非开颅蛛网膜下隙出血组(SAH组)、非开颅蛛网膜下隙出血后蜕皮甾酮(20 mg/kg,2次/d)干预组(EDS干预组)。光学显微镜观察大脑皮质病理学改变,TUNEL法检测大脑皮质神经细胞凋亡率,免疫组化分析Bcl-2,Caspase-3蛋白表达变化。结果与SAH组比较,EDS干预组脑含水量有不同程度好转,大脑皮质神经细胞凋亡减少,Bcl-2表达较SAH组增加,Caspase-3表达减少。结论蜕皮甾酮对蛛网膜下隙出血后大脑皮质损伤有保护作用,其保护作用可能是通过减轻脑水肿、抗凋亡等途径减轻神经细胞损伤。 Objective To explore the protective effect of ecdysterone on the brain cortex injury following subarachnoid hemorrhage and its mechanism. Methods Sixty Sprague-Dawley rats were divided randomly into three groups: SAH group,EDS group (20 mg/kg,twice a day) and control group. At 24 h after the treatment ,the brain water contents were measured. The pathological changes were observed un- der light microscope,the levels of Bcl-2 and Caspase-3 were measured by immunohistochemistry, and the apeptosis rate was detected by TUNEL. Results Compared with SAH group,the brain water contents decreased in EDS group,the apeptotic neuron ceils and Caspase- 3 expression also decreased significantly,while Bcl-2 increased. Conclusion EDS can relieve the brain cortex injury following SAH, which could be probably induced by relieving the neural cell injury via anti-cerebral edema and anti-apeptosis.
