蜡梅属植物叶化学成分HPLC指纹图谱的构建 被引量:9

HPLC fingerprints establishment of chemical constituents in genus Chimonanthus leaves

摘要 采用高效液相色谱法构建蜡梅属植物叶化学成分的HPLC指纹图谱,并在蜡梅属5个种间进行比较。采用Hi-bar C18色谱柱(4.6 mm×250 mm,5μm);流动相A为水溶液,B为乙腈,梯度洗脱,0~20 min,6%~20%B;20~30 min,20%~25%B;30~40 min,25%~45%B;40~50 min,45%~80%B;50~80 min,80%~85%B;80~90 min,85%~100%B;90~110min,100%B;流速0.8 mL.min-1;检测波长228 nm;柱温为30℃。构建了蜡梅属植物叶片化学成分的HPLC指纹图谱,其中5个化学成分通过对照品得到指认,5种蜡梅属植物对照指纹图谱差异明显,可进行种间辨认比较。根据该色谱条件构建的HPLC指纹图谱可以作为蜡梅属植物分类及质量评价的参考依据。 This study was aimed to establish the HPLC fingerprints of the genus Chimonanthus leaves and compare the constituents distribution among five Chimonanthus species. The analysis was conducted on a Cls column (4. 6 mm×250 mm, 5 μm) with the mobile phase containing acetonitrile-water in gradient program: acetonitrile (B), 0-20 min, 6%-20% ; 20-30 min, 20%-25% ; 30-40 min, 25%-45% ; 40-50 min, 45%-80% ; 50-80 min, 80%-85% ; 80-90 min, 85%-100% ; 90-110 min, 100%. Flow rate was 0. 8 mL · min^-1 and detection wavelength was 228 nm. Column temperature was set at 30℃. The HPLC fingerprints of the five Chimonanthus species have been established. Ch. praecox, Ch. nitens, Ch. salicifolius, Ch. Zhejiangensis and Ch. grammatus have significant difference in constituents distribution and contents. Five standard substances as common compounds were confirmed in chromatography fingerprints. The method can be used as quality evaluation and classicfication of the genus Chimonanthus.
