摘要
目的观察复方甘芍贴剂对多囊卵巢综合征(PCOS)模型大鼠卵巢组织形态和超微结构的影响。方法30只SD大鼠随机分为正常对照组、PCOS模型组、复方甘芍贴剂组,来曲唑法造模成功后应测定大鼠性激素水平;给予不同贴剂治疗后再次测定大鼠性激素水平,光镜观察大鼠卵巢组织形态改变,电子显微镜观察卵巢超微结构。结果应用来曲唑干预21d之后,PCOS模型组大鼠血清黄体生成素(LH)、卵泡刺激素(FSH)及睾酮(T)的水平明显高于对照组,而雌二醇(E2)和孕酮(P)水平低于对照组,提示造模成功。给予贴剂治疗后,与PCOS模型组比较,复方甘芍贴剂组大鼠血清LH、FSH及T水平下降,血清E2、P水平升高,差异有统计学意义。与PCOS模型组比较,复方甘芍贴剂组光镜下卵巢基本恢复为正常对照组卵巢形态,可见成熟卵泡及黄体的生成;电镜下见观察颗粒细胞凋亡所特有的超微结构变化明显改善,可见正常的颗粒细胞和及核仁,线粒体增多,有大量滑面内质网;卵泡膜细胞形态基本恢复。结论复方甘芍贴剂能够从形态学上改善PCOS大鼠的卵巢组织,恢复排卵功能。
Objective To investigate morphologic and ultrastructure changes of polycystic ovary syndrome model in rats treated with Compound Ganshao Paste. Methods Thirty female rat models of polycystic ovary syndrome were established by letrozole and then were randomly divided into three groups: control group, PCOS model group, and Compound Ganshao Paste therapy group. Hormone concentrations were examined by radioimmunoassay after the rat models were estab- lished and pathohistologic changes of rats'ovarian structure were observed by hematoxylin-eosin (H&E) staining after treated with different paste. The ultrastructure of the rats' ovary was investigated by transmission electron microscope. Resuits Compared with the control group, serum levels of luteinizing hormone ( LH), follicle stimulating hormone (FSH) and testosterone (T) were significantly increased after letrozole intervention, and serum levels of estradiol (E2) and progesterone (P) in experimental group were significantly decreased compared. After Compound Ganshao Paste intervention, serum levels of LH, FSH and T were lower and levels of E2 and P were higher when compared with the PCOS group. The ovarian tissue of Compound Ganshao Paste therapy rats showed an improvement in developmental stages of follicle and corpus luteum formation with an increase in the layers of cumulus granulosa cells when compared with the PCOS model group under optical microscope. In the Compound Ganshao Paste therapy group, ovarian granulosa cell apoptosis under microscopy showed significant improvement, including normal cumulus granulosa cells and an increase in the number of mitochondria, and a large number of smooth endoplasmic reticulum, theca cells increased significantly under the transmission electron microscope. Conclusion The Compound Ganshao Paste therapy could improve the ovarian morphology of polycystic ovary syndrome in rats and restore ovulation.
